Tuba plants is known to contain the chemical rotenone, which is have an insecticidal properties. The objectives of this study were conducted to identify the Tuba plants and quantify their rotenone contents. Materials and Methods: Nine tuba plants of different local names were collected from various locations in Peninsular Malaysia. Random Amplification of Polymorphic DNA (RAPD) and internal transcribed spacer (ITS) marker of 9 Tuba plant accessions were used to identify the species. Results: Both methods were equally adequate for Tuba plants species identification. Four different species were obtained from nine accessions and they were Derris elliptica, Paraderris elliptica, Fordia splendidissima and Paraderris piscatoria. These species are the new record in Peninsular Malaysia. All accessions contained rotenone with the concentration varying from 0.25-1.02 μg mL–1. The highest rotenone content, 1.02 μg mL–1 was from D. elliptica (Tuba merah). This indicated that the two techniques (RAPD and ITS) are equally appropriate for the analysis of genetic diversity in Tuba plants. With this purpose, 19 representativesamples were selected from the 2009-2012 year period. Pollenanalysis showed a predominance of native species of differentlandscapes such as forests and wetlands. Of the total number ofsamples, eleven were mixed or multifloral and eight samples weredominated by different native nectariferous woody species. The colourof honey, according to the Pfund scale, ranged from extra lightamber to dark amber. Total phenolic content ranged from 40.18 to118.82 mg GAE/100 g honey. Flavonoid content varied between6.94 and 67.76 mg QE/100 g honey, and antioxidant capacity, evaluatedby the DPPH radical method, between 17.36% and 56.53%.Extra light and light amber honeys had lower levels of total phenoliccompounds, total flavonoids, and antioxidant capacity than amberand dark honeys. The correlation observed between colour andflavonoids (r=0.78) was higher than that between colour and totalphenols (r=0.53). A direct relationship between phenolic contentand antioxidant capacity (r=0.91) was found. The studied honeys, ascompared to honeys from other regions, were rich in flavonoids andshow a noticeable antioxidant activity. This reduction persisted during the whole study period.Nevertheless, the woody species tended to recover their percentagecover towards the end of study. At the end of the study period, significantdifferences (P 0.05) in woody species mean density were detectedamong treatments throughout the study period. At the end of thestudy period, the foliar cover and density of desirable perennial grassspecies was higher (P 0.05) were detected in intermediate perennial grassesfoliar cover and density among treatments. Our results suggest thatthe reduction of the woody species cover might encourage increasedthe foliar cover and density of desirable perennial grass species.
