Microbes that act as probiotics are lactic acid bacteria (LAB) that can produce metabolites such as lactic acid, hydrogen peroxide, and bacteriocins. This study aimed to obtain lactic acid bacterial isolates from tape and tempeh, and to test the potential of LAB as a probiotic candidate by activity test as an antidiarrhea and its resistance to gastric pH and bile salts. The fermentation products used as a source of LAB isolates are tempeh sumber mas merk, and yellow cassava tape, sari madu merk from Jember. The results of the first stage regarding the isolation of LAB using GYP media showed that there were 2 LAB isolates (TaJ.14 and TaJ.15) from the tape and 4 LAB isolates (TeJ.18, TeJ.22, TeJ.24, and TeJ.25) from tempeh. The results of the antidiarrheal test using the disc diffusion method (oxoid) showed that TaJ.14 and TaJ.15 isolates were able to inhibit Bacillus subtilis, Escherichia coli, and Shigella dysentriae, while TeJ.18, TeJ.22, TeJ.24, TeJ.25, and Lactobacillus casei (control) was only able to inhibit B. subtilis and E. coli. The results of LAB resistance to gastric pH showed that the TeJ.25 isolate had the highest percentage of pH 3 and 2.5 resistance (51.13 and 33.03%) compared to other isolates and controls. LAB resistance test results against bile salts (oxgal) showed that the TeJ.22 isolate had the highest percentage of resistance (75.10%) compared to other isolates although was still higher in control (75.99%).","PeriodicalId":17683,"journal":{"name":"Jurnal Biodjati","volume":null,"pages":null},"PeriodicalIF":0.0,"publicationDate":"2021-11-29","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"81996502","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The promising target for drug discovery of COVID-19 was SARS-CoV-2 Main Protease (Mpro), that has been successfully crystallized along with its inhibitor. The discovery of peptide-based inhibitors may present better options than small molecules for inhibitor SARS-CoV-2 Mpro. Natural compounds have such a wide potential and still few explored, Zizyphus spina-christi is one of the medicinal plants that have many pharmacological activities and contains a peptide compound from alkaloids class, i.e. cyclopeptide alkaloids, that is interesting to explore as SARS-CoV-2 Mpro inhibitor. The compound structure was drawn and optimized using density functional theory 3-21G method. The protein chosen was the high resolution of SARS-CoV-2 MPro receptor (1.45 Å) with PDB ID: 6WNP, in complex with boceprevir. Molecular docking simulation was performed using Autodock4 with 100 numbers of GA run, the validation methods assessed by RMSD calculation. Furthermore, the prediction of pharmacological activity spectra was carried out using the PASS Prediction server. The results showed RMSD value was 1.98 Å, this docking method was valid. The binding energy of all compounds showed better results than the native ligand (Boceprevir). The in silico PASS prediction results indicated that all compounds showed antiviral activity. Some compounds showed protease inhibitory activity, i.e Ambiphibine-H, Franganine, and Mauritine-A, and the highest Pa (Predicted activity) value showed by Mauritine-A compounds. It can be concluded that the cyclopeptide compounds of Zizyphus spina-christi were indicated to have a potential as COVID-19 therapy targeting SARS-CoV-2 Mpro.","PeriodicalId":17683,"journal":{"name":"Jurnal Biodjati","volume":null,"pages":null},"PeriodicalIF":0.0,"publicationDate":"2021-06-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"81090185","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Lactic acid bacteria (LAB) can be used as an alternative to prevent diseases in fish by increasing the body's resistance. This research aimed to determine the most effective isolates of LAB that increase of the resistance of carp fingerlings to the attack of E. tarda bacteria and see which isolates can produce the highest survival. The LAB isolates used were the result of isolation from the gut of carp. This study used a Completely Randomized Design (CRD) consisting of four treatments with three replications. The fish were immersed with different LAB isolates, CcB7, CcB8, and CcB15 in the same density of 108 cells / mm3. Immersion was carried out for 30 minutes with a frequency of seven days. While during the research, two immersions were carried out before the challenge test against E. tarda bacteria. The parameters observed were the number of leukocytes, hematocrit, erythrocyte, differential leukocytes, survival rate, and clinical symptoms that appeared. The results showed that all LAB isolates used in this study could increase the body resistance of carp against the attacks of E. tarda bacteria. The LAB CcB7 isolate was the most effective for enhancing the body resistance of carp fish withthe highest increase level of  leukocyte, erythrocyte, and hematocrit were 18 ± 0.057,7 ± 0,077, and 0.26± 7.31% respectively. After being challenged with E. tarda bacteria producing mild clinical symptoms, the highest increase is in monocyte and neutrophil cells was 20 and 62% respectively, the highest reduction in lymphocytes was – 9%  and the highest survival rate was 80%. ","PeriodicalId":17683,"journal":{"name":"Jurnal Biodjati","volume":null,"pages":null},"PeriodicalIF":0.0,"publicationDate":"2020-05-30","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"74766724","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Thus, the phytoplankton community structure is a good indicator of water quality due to its sensitiveness to environmental stresses. Two newly dug ponds in Institut Teknologi Sumatera may give an opportunity to study the early colonizing stages of various freshwater communities including phytoplankton. The study attempted to determine the composition and abundance of phytoplankton. Samples were collected from two ponds (A and C) in the reservoir water of Institut Teknologi Sumatera. The content of Phosphorus (P), Nitrogen (N), and Chlorophyll-a (algae biomass) were determined. Phytoplankton had higher diversity in Pond C than Pond A in the study period, in which a total of seven taxa were found, namely Bacillariophycea, Cyanophyceae, Chlorophyceae, Conjugatophyceae, Dinophyceae, Euglenophyceae, Gymnodiniaceae. The most species abundance of both ponds was Peridinium sp. and Trachelomonas sp. The Pond C had the highest mean value of the Shannon-Wiener diversity index. The Linear mixed-effect model showed that low turbidity will result in high phytoplankton diversity. The finding of this study suggests that higher phytoplankton diversity would achieve a natural carrying capacity, and thus would serves as an indicator of ecosystem health.","PeriodicalId":17683,"journal":{"name":"Jurnal Biodjati","volume":null,"pages":null},"PeriodicalIF":0.0,"publicationDate":"2020-05-30","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"84758691","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Among the popular marine ornamental fish in the local and international markets belong to the family of Chaetodontidae. The existence of this family has been reported from several areas in Indonesia. However, no information is available in terms of their phylogenetic relationships of the fish from the south coast of West Java. The aim of this research was to asses the phylogenetic relationships among ornamental fish members of Chaetodontidae in the South Coast of West Java. Fish samples were collected from two trading sites Ujung Genteng and Taman Manalusu. Collected fish were identified using a previously published study. The evolutionary relationships among species were analyzed statistically through phylogenetic analysis based on maximum parsimony algorithm and Kimura 2- Parameter substitution model as implemented in PAUP 4.0 software. Tree branching pattern supported by 1000 bootstraps pseudo-replicates and out-group comparison. Daschyllus trimaculatus used as outgroup species. Based on the study, Chaetodontidae from the South Coast of West Java formed one monophyletic group compared to Dascylus trimaculatus with Chelmon rostratus was the basal species. Therefore research on this subject needs to be conducted.  The spectrophotometry method was used to study protease, lipase, and amylase activities in barred loach. The study used 89 fish with an average weight of 3.61 ± 0.26g, and 1.68 ± 0.21g.  The results showed that pH 7.0 was the highest protease activity. Similarly, lipase activity found  between pH 7.0 and pH 8.1. Still, the amylase activity found to be high at pH 8.1. Protease activity also found to be high in the posterior intestine than in the hepato-pancreas and anterior intestine. Lipase and amylase activities did not found differences between hepato-pancreas, anterior intestine, and posterior intestine. In conclusion, the protease, lipase, and amylase activities of barred loach found along the gastrointestinal tract in both large and small fish, which shows that barred loach can digest protein, fat, and carbohydrates in diets better.","PeriodicalId":17683,"journal":{"name":"Jurnal Biodjati","volume":null,"pages":null},"PeriodicalIF":0.0,"publicationDate":"2020-05-30","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"85339336","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
