The method used a reversed-phase C18 column and methanol-water (90+10, v/v) mobile phase. The quick, easy, cheap, effective, rugged and safe method was used for extraction of Ethofenprox from water surface, which involved the extraction of 1.55 µL sample homogeneous distribution with 10 mL water, followed by the covering of oil-absorbing paper (OAP). After twenty minutes, Ethofenprox was extracted into OAP. Then, the paper was shaked in 10 mL methanol. After concentration, the final extract was transferred to an autosampler vial for concurrent analysis by HPLC. Etofenprox in a good linear relationship in the ranges was studied, 0.1–20.0 mg L−1, with high correlation coefficients (0.9999). The RSDs were 0.12%. The average recoveries of Ethofenprox fortified at different levels were within 90.9–96.5%, and the precision of the test method was 1.6–4.4%. The concentration of total proteins was fixed at 5% (w/v), and the ratio of SPI to gelatin was fixed at 4∶1 (w/w). The cross-linking of the proteins was carried out as fellows: enzyme addition level at 20U/g proteins, reaction time of 2 h and temperature of 40°C. Methods According to orthogonal experiment design L<inf>25</inf>(5<sup>6</sup>) of enzyme, osmotic pressure stabilizer(D-Mannitol), pH, enzymolysis time, we got the optimal isolation condition by the number of protoplasts. Results The optimal isolation condition is showed in results: enzymolysis time 2h, pH 5.5, D-Mannitol 0.6 mol/L, 1.5% cellulose and 0.1% pectolyase and 0.1%BSA, temperature 28°C. It also means that optimal isolation condition is D<inf>5</inf>B<inf>3</inf>C<inf>4</inf>A<inf>2</inf>. Yields, water use efficiency (WUE) and benefit of winter wheat under different water and fertilizer couplings were studied. The results showed that: the yield, WUE and benefit of winter wheat are the highest under the medium water and fertilizer. Over-fertilization affected the winter wheat yield; irrigation at jointing and seed filling stage made plants grow better, while moderate water shortage in the later growth stage have a certain role in promoting the yield formation.
