The single-shot localization is based on complete outer-volume suppression which is achieved with optimized, adiabatic BIR-4 pulses in combination with<em>B</em><sub>0</sub>gradient modulation.<em>B</em><sub>1</sub>-insensitive water suppression is achieved with BISTRO, a train of selective adiabatic full-passage pulses interleaved with<em>B</em><sub>0</sub>gradient crushers. It is based on the pulsed-field-gradient technique and relies on the fact that extracellular water diffuses freely while intracellular water is completely restricted by the relatively impermeable cell wall of the bacterium. At high wave vectors, the signal from extracellular water is completely suppressed while the signal from intracellular water is comparatively unaffected. This technique has been applied to the mapping of bacterial distributions in porous media. Dietze ,&nbsp;Otto Lutz","doi":"10.1006/jmrb.1996.0150","DOIUrl":"10.1006/jmrb.1996.0150","url":null,"abstract":"<div><p>Proton-decoupled<sup>31</sup>P NMR spectroscopy was performed<em>in vitro</em>and<em>in vivo</em>in humans to investigate the temperature dependence of the chemical-shift separation δ<sub>αβ</sub>between the α- and β-phosphorus resonances and the<sup>31</sup>P–<sup>31</sup>P coupling constants of ATP using a 1.5 T whole-body imager.<em>In vitro</em>in the presence of magnesium, a linear temperature dependence of δ<sub>αβ</sub>was found that varied systematically with Mg-to-ATP ratio (<em>R</em>). This needs to be considered in studies where δ<sub>αβ</sub>is used to determine temperature. A second important point is the temperature dependence of −0.0128 ± 0.0002 ppm/°C (mean ± SD,<em>n</em>= 6) of the chemical-shift limit δ<sup>MgATP</sup><sub>αβ</sub>(<em>T</em>) of mostly mono-magnesium-complexed ATP found at<em>R</em>= 2.2 ± 0.2. This parameter is used in combination with the temperature dependences of the uncomplexed limit δ<sup>ATP</sup><sub>αβ</sub>(<em>T</em>) and the dissociation constant<em>K</em><sup>MgATP</sup><sub>D</sub>(<em>T</em>) for the determination of [Mg<sup>2+</sup><sub>free</sub>] at different temperatures. Preliminary studies<em>in vivo</em>show that δ<sub>αβ</sub>also decreases in human skeletal muscle with increasing temperature, similarly to the<em>in vitro</em>results.
