Soni, Preeti Sharma","doi":"10.26438/ijsrbs/v6i2.7480","DOIUrl":"https://doi.org/10.26438/ijsrbs/v6i2.7480","url":null,"abstract":"Chia seeds from Salvia hispanica and Basil seeds from Ocimum basilicum are increasingly being categorized as a novel food and have received scientific attention due to presence of high amounts of nutraceutical compounds. Recent studies demonstrate that both seeds are used as a medicine for the treatment of various kinds of diseases. Present study aims to determine their antihypertensive potential, anti-inflammatory action, and cytotoxicity. Hypertension is a major risk factor and Angiotensin Converting Enzyme (ACE) is one of the major regulators of blood pressure. Inflammation is response of body to an injury or infection. Drugs which are used as ACE inhibitors and anti-inflammatory possess undesirable side effects, while natural inhibitors have no side effects and are potential nutraceutical. In vitro assays were used to determine ACE inhibition and anti-inflammatory activity. The cytotoxicity assay for crude extract of defatted chia and basil seeds was done on human histiocytic lymphoma cell line (U937) by MTS assay. ACE inhibition activity assay showed that ethanolic and hexane extracts of both seeds have distinct ACE inhibitory activity. Both seed extracts had shown higher activity at 150 and 50 μg/mL concentrations respectively. Results of anti-inflammatory assay showed maximum protection of HBRC (Human Red Blood Cell Membrane Stabilization Method) at 50 and 75 μg/mL concentrations for both seeds in ethanolic and methanolic extracts. MTS assay showed 73.20% and 70.85% cytotoxicity of Chia and Basil seeds at 12.50 and 50μg/mL concentrations respectively. IC50 values of Chia and Basil seeds were found to be 14.09μg/mL and 23.46μg/mL. This study provides direction for future investigation concerning detailed assessment of therapeutic potential of Chia and Basil seeds. The Ethonoloic and Choloroform leaf extracts of G.sylvestre showed significant antibacterial against some clinically important gram negative and gram positive bacteria such as Staphylococcus aureus (ATCC6538), Enterococcuc faecalis (ATCC 29212) and Escheria coli (ATCC8739), Salmonella enterica (ATCC 10798) respectively. The in-vitro antibacterial activity was performed by agar well diffusion method (25, 50, 75 and 100 μg/ml ). The maximum inhibition zone of ethonolic leaf extract was found in 100 μg /ml against gram negative bacteria such as Salmonella enterica (26mm), Escheria coli (24mm) and gram positive bacteria such as Enterocococcus faecalis (26mm) and Staphylococcus aureus (24mm). The maximum inhibition zone of chloroform leaf extracts found in 100 μg/ml (25,50,75, and 100 μg/ ml) against gram negative bacteria such as Salmonella enterica (24mm), Escheria coli (24mm) and gram positive bacteria Enterocococcus faecalis (23mm) and Staphylococcus aureus (23mm). The phytochemical tests were carried out and showed that the antibacterial activity of plant Gymnema sylvestre leaves were due to presence of phytochemical compounds such as Alkaloids, Flvanoids, Glycosides, Reducing sugar, Tannins, Saponins, Polysaccharides and Phenols. The ethonolic and choloroform leaf extracts of Gymnema sylvestre showed maximum inhibition against Staphylococcus aureus, Enterocococcus faecalis, Salmonella enterica and Escheriacoli. The Ciprofloxacin was used as a positive control. The leaves of Justicia gendarussa were collected from Kadukaval in January 2018 Thanjavur, Tamil Nadu, India. The powder leaf was extracted with aqueous and 70% methanol for 24 hours and studied for FTIR, HPLC and NMR. A preliminary phytochemical testing of the leaves extract where than to identify the phytoconstiuents, which reveals that presence of tannin, saponin, flavonoids, steroids, terpenoids, triterpenoid, carbohydrate, anthroquinone, polyphenol, glycoside bioactive compound was isolated by column chromatography technique. The collected flavonoid fractions was purified by thin layer chromatography. FTIR, HPLC and NMR studies were carried out to find the structure of bioactive compound Quercetin. 1 HNMR and 13 C – NMR that reveals the structure of flavonoids. The compound was identified as 3, 3’, 4’, 5, 7 – pentahydroxyflavanone by 1 HNMR and 13 C – NMR. All these data obtained in the present investigation supported the rich source of phytochemicals preent in J.gendarussa leaves extract and therby traditional claim associated with J. gendarussa literature.
