Less attention has been paid to the κ opioid peptide (KOP) system, likely because drugs that activate the KOP system have low abuse potential. Indeed, activation of the KOP system often is reported to have effects on brain and behavior that are opposite to MOP activation. The literature suggests that dynorphin and its κ receptor are, indeed, unique among the opioids. That uniqueness has generated interest in our laboratory. We have taken a different approach to understanding the KOP system. Rather than focusing on drug abuse, we have adopted mood and anxiety as a model for investigation of the KOP system. The assumption is that clarification of the underlying mechanisms and behavioral outcomes of κ agonists and antagonists will help inform the KOP system for other situations, including drug use and abuse. Distribution of neurolectin-binding receptors glycoproteins and glycolipids in the same structures, their role in the processes of migration of neurons and other mechanisms are studied as well. Two membrane neurolectins VL-1 and VL-NAGA have been revealed in synaptosomes, which according to our hypothesis are actively involved in absorption and secretion of neurotransmitters. It was established that VL-1, located on the outer membrane of synaptosomes participates in absorption and metabolism of biologically active substances, though VL-NAGA, which is located in the inner side of the membrane is also involved in their metabolism. It is considered that negative effects of wheat germ agglutinin (WGA) lectins, among them neurotoxic, cardiotoxic and immunotoxic, can be avoided by means of well toasted bread, because in the process of toasting lectins undergo denaturation and lose their toxic activity. Food should be chewed well, until it becomes saturated with glycoprotein mucine, which is component of saliva, as N-acetyl-D-glucosamine binding and inactivating WGA is presented in its active centre. Hence addition of antioxidants/phytochemicals is recommended. In the present study, we have used fish oil (FO) in combination with Bacopa monnieri (BM) and examined the neuroprotective propensity of the combination (FO+BM) employing chronic 3-nitropropionic acid (NPA) model of neurotoxicity in rats. MATERIALS AND METHODS: Growing male rats administered NPA (10 mg/kg bw, 14d) were supplemented with FO (2 mL/kg bw), BM (100 mg/kg bw) or combination (FO+BM) for 14 days. RESULTS: Higher degree of protection was discernible among FO+BM rats against NPA induced behavioral alterations. Further, NPA administration resulted in significantly elevated oxidative markers in brain regions (striatum- St, cerebellum- Cb) while robust protection was evident among rats supplemented with FO+BM. Depleted GSH levels in St were restored among all the supplemented groups. The combination also predominantly normalized the activity of antioxidant enzymes in brain regions. Further, significantly depleted dopamine levels in St were partially restored only among FO+BM rats. However, the combination did not offer protection against NPA induced mitochondrial dysfunction. Volta, Ommega Internationals","doi":"10.15436/2377-1348.17.1630","DOIUrl":"https://doi.org/10.15436/2377-1348.17.1630","url":null,"abstract":"Objective: The study aim is to verify whether treatment with a new combination of tanacethum partenium, 5-hydrossitriptophan (5-http) and magnesium (Aurastop®) reduces headache frequency and intensity in patients suffering from episodic migraine without aura when used in migraine prevention. Methods: Forty patients, suffering from migraine without aura for at least 6 months with monthly frequency of 3 to 8 crises and presence of headache of 4 to12 days, were enrolled in this open study and treated orally with Aurastop twice daily for 3 months. The primary endpoint was reduction of migraine frequency (headache days per month) over an observation period of 3 months. The secondary endpoint was a composite of monthly frequency and intensity of pain crises, analgesics use (number of medications) and subjective change of pain intensity. Results: All the parameters significantly improved at the end of treatment with Aurastop. We observed a significant reduction of the number of headache days (from 8.8 ± 2.0 before treatment to 2.7 ± 1.7 post treatment, p < 0.001), as well as of the number of attacks (from 5.0 ± 1.2 per month to 2.1 ± 0.9 per month, p < 0.001), of pain intensity (from Visual Analogic Scale [VAS] 6.9 ± 1.0 to 3.3 ± 1.5, p < 0.001), and of the number of analgesics assumed by each subject (from 8.5 ± 1.6 per month to 2.4 ± 1.5 per month, p < 0.001). No serious adverse events were observed. Participants: Seventeen patients with PDC having an average of 328.8 days post-injury and eleven control participants. Main measures: All participants underwent two DTI studies. Fractional anisotropy (FA) values were analyzed using whole-brain analysis (WBA) of 7 patients with no marked brain deformities; mean FA values of forceps minor (mFAFM) were subsequently measured by regional-brain analysis (RBA). Relationships between WB and regional FA values and recovery from PDC were evaluated. Results: WBA in the PDC group showed that FA values were significantly lower in multiple white matter regions (P < 0.05), particularly in part of the forceps minor, in the second scan than they were in the first scan. RBA showed that mFAFM significantly decreased (P < 0.01). Further, WBA revealed that a significant positive correlation was observed between the degree of recovery from PDC and the difference in the number of voxels with FA values > 0.2 (r = 0.65, P < 0.01) between the first and second scans.
