Since experimental studies have confirmed the presence of cutaneous H2 receptors partially conflicting results have been reported on the use of H2 antagonists in c./c.r.U. 20 patients with c./c.r.U. of idiopathic type were treated with cimetidine plus clemastine or placebo plus clemastine in a double-blind crossover study. Before this treatment traditional H1 antagonists alone failed to show any satisfying therapeutic effect. The results reveal that combined therapy is statistically more effective than corresponding H1 antagonists alone (P = 0.001). In the present study, however, 36 psoriasis blood sera were examined before and fourteen days after the coal tar and dithranol (anthralin, cignolin) therapy onset to determine D and E fibrinogen fragments. These serum levels were compared with those in 36 blood donors. For such investigations, the knowledge about the behaviour of the test system under standardized conditions is an important prerequisite. In the 72 h short-time-culture, no changes in T-lymphocytes were found. Small decreases occurred in the subpopulations of cytotoxical/suppressor cells, and inducer/helper cells, respectively. Number of cells expressing class II antigens of the main histocompatibility complex decreases, too (to 62%). A relief of joint pain could be detected under this treatment both after 6 weeks (improvement of the Ritchie-indices: 44%) and after 12 months (improvement: 28%). In contrast to this, X-ray investigations show a worsening of the disease after 12 months. The present study provides evidence that a subpopulation of human keratinocytes most probably at a certain stage of differentiation, possesses the mouse erythrocyte binding receptor the marker of a human B lymphocyte subset. {"title":"[Effect of pepstatin A on Candida albicans infection in the mouse].","authors":"C Zotter, U F Haustein, C Schönborn, H D Grimmecke, H Wand","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>The intravenous injection of 10(6) to 10(7) Candida albicans cells revealed to be a reliable model in mice produce infections of the kidney. Higher germ contents could be yielded in the kidney after the application of protease positive Candida-strains as compared to protease negative ones. Additionally, after the infection with protease positive strains a proteolytic activity could be found in the kidney homogenates in vitro on casein plates. The modulation of this Candida infection by pepstatin A, an inhibitor of extracellular yeast proteases in vitro, has also been studied in the same mice model in vivo. The growth rate of Candida albicans has been measured in the left kidney by counting the germ content as described by Haenel. Infections could be reduced after single doses of 120 micrograms pepstatin A contrary to 60 micrograms pepstatin A. The same was with three doses of 180 micrograms at 24 h intervals. This protease inhibitory effect could also be found in kidney homogenates in vitro on casein plates and lasted until 48 h post injection.
