In contrast, science requires universality from us. However, until the end of 20th century, there was no such a common forum for chromosome science in Asia. In 1998, while the 18th International Congress of Genetics was being held in Beijing, China, chromosome researchers attending from Japan (Dr. Kiichi Fukui), China (Dr. Youzhi Ma), Korea (Prof. Jae-Wook Bang) and Th ailand (Dr. Somsac Apisitwanich) discussed the possibility of creating an Asian forum for chromosome researchers. An enthusiastic discussion followed and unanimously concluded with the decision to have the first Asian Chromosome Colloquium (ACC) at Beijing. One of the reasons why an ACC was agreed upon is that there was already a long tradition of bilateral conferences between China and Japan initiated in 1987 at Beijing (Hong 1989) and in 1992 at Tianjin (Tanaka 1992) by Profs. Hao Shui (Northeast Normal Univ.), Chen Ruiyang, (Tianjin Univ.), Hong Deyuan (Inst. Botany, Academia Sinica) Ryuso Tanaka, Kenji Taniguchi, and Masashi Nakata (Hiroshima Univ.). Tsuchiya","doi":"10.11352/SCR.17.3","DOIUrl":"https://doi.org/10.11352/SCR.17.3","url":null,"abstract":"5-azadeoxycytidine (5-aza-dC)-induced undercondensation (UC) was described with cultured lung fibroblasts from three mammalian species, Pipistrellus abramus , Millardia meltada , and Apodemus argenteus , all of which carry a large amount of C-heterochromatin (C-block) on their X chromosomes. The C-blocks were GC-rich in P. abramus , AT/GC-balanced in M. meltada , and AT-rich in A. argenteus , as revealed by fluorescent staining with quinacrine mustard (QM) and chromomycin A 3 (CMA). P. abramus conspicuously undercondensed, irrespective of autosomes or gonosomes, in all C-band regions including C-blocks. The extent of UC was most prominent in P. abramus , rather weak in M. meltada , and weakest in A. argenteus , suggesting a close relation to the amount of GC base pairs contained in C-heterochromatin. Thus 5-aza-dC treatment may be an easy method for detect-ing GC-rich heterochromatin. The appropriate conditions for inducing UC in the C-blocks were 10 -5 –10 -6 M and 2–3 hours. The decondensing effect of 5-aza-dC on the C-blocks was time- and concentration-dependent in all species examined, asynchronously acting with certain time lag between the early-replicating X and late-repli-cating X. Several studies have identified the distribution and function of RNA molecules in mitosis; however, the relationships between RNA molecules and condensed chromosomes during mitosis have not been systematically studied. In this study, we investigat-ed specific RNA molecules during mitosis and identified structural components that surround the chromosomes, called Mitotic Chromosome Coating Spheres (MiCCS), by using an in-cell deproteinization assay. Furthermore, we observed the unique distribution patterns of the RNA molecules—in terms of mitotic chromosomes, MiCCS, mitotic cytoplasm, and outer membranes of mitotic cells—by using a mitotic three-dimensional RNA fluorescent in situ hybridization (3D-RNA-FISH).
