This study investigated the spatial distribution and molecular expression of estrogen receptor alpha (ERα), and prostaglandin E2 receptor subtype 2 (EP2) and 4 (EP4) in the caprine cervix during the follicular, luteal, and early pregnancy stages. Cervical tissues from does were categorized into these three stages. The immunolocalization and protein expression of ERα, EP2, and EP4 were assessed using immunohistochemistry and Western blot analysis. Additionally, the relative mRNA expression of ESR1, PTGER2, and PTGER4 was evaluated by RT-qPCR. Results demonstrated a synchronized ERα-EP4 signaling axis during the follicular stage, with peak overall protein expression in the epithelium and smooth muscle. Post-ovulation, ERα and EP4 expression significantly declined, whereas epithelial EP2 was observed to be abundant during the luteal phase. During early pregnancy, ERα and EP4 protein levels dropped to their lowest points. Notably, a major disconnect occurred between gene activity and protein production; while the PTGER4 gene was highly active, EP4 protein remained minimal. Within the receptors examined, these findings indicate that the caprine cervix regulates ERα, EP2, and EP4 in a compartment-specific manner across the reproductive cycle. The combination of ERα and EP4 prepares the tissue for mating, whereas EP2 secures the barrier after ovulation. During early pregnancy, inhibiting the synthesis of EP4 protein serves as a crucial mechanism to avoid premature cervical dilation.</p>","PeriodicalId":94190,"journal":{"name":"Reproductive biology","volume":" ","pages":"101294"},"PeriodicalIF":0.0,"publicationDate":"2026-10-15","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"149198590","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

This study utilized Four-dimensional independent data acquisition (4D-DIA) high-throughput quantitative proteomics to systematically analyze the global proteomic changes in healthy human sperm before and after in vitro capacitation. A total of 5812 quantifiable proteins were identified, of which 201 were significantly differentially expressed (Fold Change > 1.5, P < 0.05). Functional analysis revealed that upregulated proteins were highly enriched in core energy metabolism pathways, such as oxidative phosphorylation and the mitochondrial respiratory chain, confirming the driving role of an \"energy burst\" in capacitation. Furthermore, processes including RNA processing and translation, as well as post-translational modifications, were significantly activated, and the interaction network demonstrated a tight coupling between energy metabolism and translational machinery components. Concurrently, this study identified a group of potential novel regulators (e.g., TMEM89, TEX29) with high testis-specific expression that are upregulated post-capacitation. This study provides the first systematic mapping of the multidimensional proteomic dynamics during sperm capacitation, offering novel molecular biomarkers and potential targets for the etiological diagnosis of male infertility and assisted reproductive interventions.</p>","PeriodicalId":94190,"journal":{"name":"Reproductive biology","volume":" ","pages":"101290"},"PeriodicalIF":0.0,"publicationDate":"2026-10-15","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"149198673","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
