{"title":"Organized spirochetal behavior in human subgingival plaques - A virulence factor in periodontal infections?","authors":"Paul H Keyes, Thomas E Rams","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>The organization and behavior of spirochetes in human subgingival plaques was studied with phase-contrast microscopy. Wet-mounts of non-dispersed subgingival microbial specimens from deep pockets of 10 persons with untreated adult periodontitis revealed \"brush formations\" with outer coatings of closely-massed spirochetes exhibiting synchronized motility. Monolayers of closely-packed spirochetes co-aggregated with \"brush formation\" monofilaments were obtained by using mineral oil as a mounting medium for wet-mount preparations. Spirochetes were observed to produce collectively coordinated metachronal wave patterns with their cell movements along the outer surfaces of the \"brush formation\" monofilaments, rather than flexing independently and at random. {"title":"Influence of diet and spiramycin on <i>Actinomyces viscosus</i>-associated experimental periodontitis.","authors":"Paul H Keyes, Thomas E Rams, Harold V Jordan","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>The effect of two diets, with and without the antibiotic spiramycin, on <i>Actinomyces viscosus</i>-induced experimental periodontitis was studied in albino hamsters. Severe periodontitis and root caries developed in <i>A. viscosus</i>-infected hamsters fed a high sucrose diet. Hamsters heavily infected with <i>A. viscosus</i> but fed a finely powdered laboratory chow comprised of whole grains and oats formed discrete marginal plaques, but did not develop periodontitis. <i>A. viscosus</i> was not recovered from the plaque microflora of animals receiving spiramycin, and periodontal pathosis did not occur. {"title":"The influence of puberty on spirochetal numbers assessed by dark field microscopy: a pilot study.","authors":"S Zissopoulos, M Midda","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>There seems to be a difference in the susceptibility, prevalence and severity of periodontal disease between children and adults. These differences could be due to; composition of plaque, host response or tissue alterations occurring with age. One of the micro-organisms suspected to play a role in the initiation of periodontal disease is the spirochete. A quick and simple way to observe spirochetes is via dark-field microscopy. The present study sample subgingival plaque from the crevices of 6 prepubertal and 6 postpubertal children and observed it under darkfield microscopy. The technique did not involve dispersion. The results showed a higher percentage of spirochetes for the postpubertal group (15.19% in the prepubertal and 21.62% in the postpubertal) but statistical analysis failed to show any significant difference. {"title":"The effects of various agents on root surfaces (a scanning electron microscopy study).","authors":"K Balos, B Bal, K Eren","doi":"","DOIUrl":"","url":null,"abstract":"<p><p>In periodontitis, root surface alterations have been observed in the cementum due to the presence of toxic substances mainly endotoxins of various microorganisms. Elimination of endotoxins from diseased root surfaces by root planing and demineralisation procedures seem to be essential for new attachment procedures. In this study the effects of citric acid and tetracycline HCl on periodontally involved root surfaces which had been scaled and root planed were evaluated by scanning electron microscopy. Our results showed that scaled, root planed and citric acid treated surfaces were relatively even with many dentinal tubules.
