This study introduces an innovative therapeutic platform that combines transarterial embolization (TAE) with ferroptosis induction for enhanced HCC treatment. We developed monodisperse erastin-loaded microspheres using a microfluidic technique, with a biodegradable PLA-PEG-PLA copolymer as the matrix material. By adjusting the flow rate and concentration of the continuous and disperse phases, the particle size and distribution were precisely controlled. Importantly, drug release kinetics and antitumor efficacy were effectively regulated by modulating the hydrophobic segment molecular weight of the copolymer, with lower molecular weights PLA chains facilitating faster release and superior therapeutic outcomes. In vitro and in vivo evaluations demonstrated that erastin-loaded microspheres significantly inhibited tumor growth through ferroptosis induction, outperforming bare microspheres in a VX2 rabbit model. The increased efficacy is due to the combined effects of vascular embolization and ferroptosis. This work establishes a novel treatment paradigm that integrates material design with molecular mechanism for effective interventional therapy, offering a promising translational strategy for HCC management.","PeriodicalId":65723,"journal":{"name":"现代材料动态","volume":"1 1","pages":"100007-100007"},"PeriodicalIF":0.0,"publicationDate":"2026-01-12","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147884848","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Here, we report a magnetic nanosystem VPNS@PEI-Fe3O4-Apt based on functionalized two-dimensional violet phosphorus nanosheets (VPNS) by iron oxide magnetic nanoparticles (Fe3O4), and bacteria-specific recognition aptamers (Apt) for early sepsis diagnosis and complete in vitro blood disinfection. We demonstrated that the VPNS@PEI-Fe3O4-Apt nanosystem can achieve the identification and enrichment of bacterial species in blood, resulting in the successful diagnosis of sepsis caused by a single bacterium (Staphylococcus aureus) or multiple bacteria (Staphylococcus aureus and Escherichia coli). The strategy of VPNS@PEI-Fe3O4-Apt nanosystem has high detection sensitivity (~10 colony-forming units), while significantly reducing diagnostic turnaround time to within 2 h, suggesting that it has great potential for diagnosing early sepsis in clinical therapy. Furthermore, owing to the antibacterial ability derived from the excellent photodynamic (PDT) effect of VPNS and its good biocompatibility, efficient and safe complete disinfection of blood in vitro is achieved. We believe that the VPNS@PEI-Fe3O4-Apt magnetic nanosystem provides a new strategy for bacterial detection and treatment of sepsis.","PeriodicalId":65723,"journal":{"name":"现代材料动态","volume":"1 1","pages":"16-26"},"PeriodicalIF":0.0,"publicationDate":"2025-09-16","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://media.sciltp.com/articles/2509001245/2509001245.pdf","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147902290","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

However, light disinfection primarily relies on reactive oxygen species (ROS), which are nonselective, leading to undesired damage to normal cells. Therefore, developing a photosensitizer that can selectively kill bacteria and protect normal cells is highly desirable based on the structural difference between normal cells and bacteria. In this study, a bacterial redox potential-boosted photocatalytic concept is proposed. Here, we designed a selectively bacteria-killing photosensitizer, hydroxyapatite/MoS2 coating in Ti6 implant (HA/MoS2-Ti6), which could accept the electrons from bacteria due to the potential difference between HA/MoS2 and the bacterial outer membrane. Then it further boosted the separation of electrons and holes of HA/MoS2 produced by 660 nm light irradiation, which enhanced the photocatalytic activity of HA/MoS2-Ti6. Density functional theory calculation further demonstrated the complete electron transfer circulation between HA/MoS2-Ti6 and the bacterial outer membrane. The produced ROS and changed electrons transfer pathway altered bacterial membrane potential and intracellular ROS, leading to bacterial death (92.99 ± 0.84% against Staphylococcus aureus and 94.70 ± 3.60% against Escherichia coli). Meanwhile, HA/MoS2-Ti6 has great biocompatibility with Raw 264.7, L929, and MC3T3-E1 with/without light irradiation. HA/MoS2-Ti6 can also enhance MC3T3-E1 into osteoblasts due to the osteoinduction of HA. HA/MoS2-Ti6 induced macrophages to differentiate into M2 under light irradiation. The strategy-based nano-bio interface offered a new platform to clear microbes and enhance cell differentiation simultaneously.","PeriodicalId":65723,"journal":{"name":"现代材料动态","volume":"1 1","pages":"1-15"},"PeriodicalIF":0.0,"publicationDate":"2025-07-22","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"https://media.sciltp.com/articles/2507000989/2507000989.pdf","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147903366","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
