In this study, we investigated the preventiveneffects of nobiletin (NOB) on a mouse model of concanavalin A (ConA)-inducednacute liver injury. We observed that NOB (10 mg/kg) pretreatment ofnConA-treated mice significantly lowered the levels of liver enzymesnincluding alanine aminotransferase (ALT) and aspartate aminotransferasen(AST), decreased the intracellular generation of reactive oxygen speciesn(ROS), and suppressed the release of inflammatory cytokines such asntumor necrosis factor-α (TNF-α) and interferon-γn(IFN-γ). Pathological data suggested that pretreatment withnNOB ameliorated ConA-induced liver damage by promoting proliferationnand alleviating apoptosis of hepatocytes. Furthermore, significantnsuppression of the c-Jun-activating kinase (JNK) signal was also observednin NOB-pretreated liver tissues compared with that of ConA treatmentnonly. In addition, an in vitro mechanism study confirmednthat the addition of NOB protected hepatocytes via inhibition of JNKnactivation, manifesting that alleviation of JNK-induced apoptosisnof hepatocytes is correlated with NOB protection in acute liver injury.","PeriodicalId":523789,"journal":{"name":"Figshare","volume":"1 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2020-06-14","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147921983","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
