To understand the molecularnmechanisms of silk production, the posterior silk glands aged to thenthird day of the fifth instar were analyzed from the ZB strain withnlow silk production and from the control strain Lan10. Using isobaricntags for relative and absolute quantification (iTRAQ) quantitativenshotgun proteomics and RNA-sequencing-based transcriptomics, 139 proteinsnand 630 transcripts were identified as novel in the ZB strain comparednwith the Lan10 strain, indicating that these results significantlynexpand the coverage of proteins and transcripts of the posterior silknglands in the silkworm. Of the 89 differently changed proteins, 23nwere increased, and 66 were decreased. Of the 788 transcripts, 779nwere upregulated, and 9 were downregulated. These results confirmnthat decreased energy utilization/protein translation and enhancednprotein degradation are the key factors in lower silk production.nMoreover, this study provides novel insight into the molecular changesnthat may result in lower silk production, namely, a combination ofnimpaired transcription activity, missed protein folding/transport,nand lowered yields of the main components of fibroin, along with weakenedngrowth/development of the posterior silk gland.","PeriodicalId":523629,"journal":{"name":"Figshare","volume":"1 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2016-02-12","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147919263","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
