The aim of this study was to examine its influence on cholesterolnefflux and to address the underlying mechanism of action. Leoliginnincreases apo A1- as well as 1% human plasma-mediated cholesterolnefflux in THP-1 macrophages without affecting cell viability as determinednby resazurin conversion. Western blot analysis revealed that the proteinnlevels of the cholesterol efflux transporters ABCA1 and ABCG1 werenupregulated, whereas the SR-B1 protein level remained unchanged uponntreatment with leoligin (10 μM, 24 h). Quantitative reversentranscription PCR further uncovered that leoligin also increased ABCA1nand ABCG1 mRNA levels without affecting the half-life of the two mRNAsnin the presence of actinomycin D, a transcription inhibitor. Proteomenanalysis revealed the modulation of protein expression fingerprintnin the presence of leoligin. Taken together, these results suggestnthat leoligin induces cholesterol efflux in THP-1-derived macrophagesnby upregulating ABCA1 and ABCG1 expression. This novel activity suggestsnleoligin as a promising candidate for further studies addressing anpossible preventive or therapeutic application in the context of atherosclerosis.","PeriodicalId":523357,"journal":{"name":"Figshare","volume":"1 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2016-06-20","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147914614","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
