This study aimednto explore the differential regulation between adipogenic and myogenicnprecursors by comparative analysis of their global proteome expressionnprofile. Adipogenic and myogenic precursors isolated from neonatalnporcine <i>longissimus dorsi</i> muscle by the preplatenmethod were verified for their unique and distinct differentiationnpotential under myogenic or adipogenic induction. A total of 433 differentiallynexpressed proteins (DEP) (<i>P</i> < 0.05 and FC >n1.20nor <0.83) between adipogenic and myogenic precursors were detectednvia a tandem mass tag (TMT)-coupled LC-MS/MS approach, including 339nup-regulated and 94 down-regulated proteins in myogenic precursorsncompared with adipogenic precursors. On the basis of functional annotationnand enrichment analysis of 433 DEP, adipogenic and myogenic precursorsnshowed significantly different metabolic pattern of energy substancesnand differential regulations of gene expression, cell structure andndevelopment, ion homeostasis, and cell motility and migration. Threenpathways including PPAR signaling pathway, phosphatidylinositol signalingnsystem, and autophagy signaling pathway, which was differentiallynregulated between adipogenic and myogenic precursors, was also discoverednto play crucial roles in cell differentiation. In conclusion, thesendifferentiated regulation patterns between the two cell subsets ofnmesenchymal precursors together defines their differentiation potentialnspecificity.","PeriodicalId":522848,"journal":{"name":"Figshare","volume":"1 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2019-01-25","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147904587","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
