The therapeutic gene is complexed by polyethyleniminen(PEI) and poly-l-glutamate (PLG), further in situ tightenednby aldehyde modified polyethylene glycol (PEG) via Schiff base reaction.nThe generated Schiff base bonds are stable in neutral pH but cleavablenin tumor extracellular pH. This gene delivery system possesses followingnfavorable properties: (1) the tunable gene delivery system is constructednby chemical bench-free “green” and fast process whichnis favored by clinician, (2) PEG cross-linking shields the surfacenpositive charges and tightens the complex particles, leading to decreasedncytotoxicity, improved stability, and prolonged circulation, (3) PEGnshielding can be rapidly peeled off by acidic pH as soon as arrivingntumors, (4) dual charge/size ultrasensitively rebounding to highernpositive potential and bigger size enhances tumor cell uptake efficiency.nA series of experiments both in vitro and in vivo are carried outnto investigate this gene delivery system in detail. An antiangiogenesisntherapeutic gene is carried for the treatment of CT26 tumors in mice,nachieving superior antitumor efficacy which is well proved by sufficientnbiological evidence. The system has great potentials for cancer therapynin the future.","PeriodicalId":522836,"journal":{"name":"Figshare","volume":"1 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2016-11-04","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147904470","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
