Allicin, fromncrushed garlic cloves, exhibits strong biological activities. In thencurrent study, the protection mechanisms of allicin against AA-causednnephrotoxicity were comprehensively examined using an <i>in vivo</i> rat model based on previous research that allicin plays a key rolenin improving renal function. The results showed that allicin attenuatednhistological changes of the kidney and ameliorated renal function.nDamaged mitochondrial structures, upregulated voltage-dependent anionnchannel 1 expression, and decreased membrane potential and adenosinen5′-triphosphate levels were observed after AA treatment. Surprisingly,nallicin notably reversed the adverse effects. Further, allicin effectivelynrestored mitochondrial function via modulating mitochondrial biogenesisnand dynamics, which might be associated with the upregulated expressionnof sirtuin 1 (SIRT1). Meanwhile, allicin dramatically activated thenSIRT1 activity and subsequently inhibited p53 acetylation, preventednthe translocation of cytochrome <i>c</i> to the cytoplasm,nand reduced the caspase expression, thus further inhibiting mitochondrialnapoptosis caused by AA. In summary, the relieving effect of allicinnon AA-caused nephrotoxicity lies in its inhibition of mitochondrialndysfunction and mitochondrial apoptosis.","PeriodicalId":522767,"journal":{"name":"Figshare","volume":"1 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2023-10-13","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147903237","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
