Microglia,nthe brain immune cells, respond differently to various α-synucleinnstrains, influencing their activation and release of harmful molecules,nleading to neuronal death. Post-translational modifications, suchnas glycation in α-synuclein, add a layer of complexity to microglialnactivation. This study aimed to explore the impact of glycation onnα-synuclein aggregation and microglial responses, which havennot been studied before. Biophysical analyses revealed that glycatednα-synuclein oligomers had distinct morphologies with a morennegative and hydrophobic surface, preventing fibril formation andninterfering with membrane interactions. Notably, there was increasedncytosolic Ca<sup>2+</sup> dysregulation, redox stress, and mitochondrialninstability compared to cells exposed to unmodified α-synucleinnoligomers. Additionally, glycated α-synuclein oligomers exhibitednimpaired binding to Toll-like receptor 2, compromising downstreamnsignaling. Surprisingly, these oligomers promoted TLR4 endocytosisnand degradation. In our experiments with oligomers, glycated α-synucleinnoligomers preferred NLRP3 inflammasome-mediated neuroinflammation,ncontributing differently from unmodified α-synuclein oligomers.nIn summary, this study unveils the mechanism underlying the effectnof glycation on α-synuclein oligomers and highlights the conformation-specificnmicroglial responses toward extracellular α-synuclein.","PeriodicalId":522697,"journal":{"name":"Figshare","volume":"1 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"1753-01-01","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147901911","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
