Tannic acid (TA), a natural polyphenolnin foods, fruits, and plants, exhibits a variety of bioactive functions.nIn our research, we decide to explore the pharmacological mechanismnof TA against HBV replication. Our results showed that TA effectivelynreduced the content of HBV DNA and viral antigens (HBsAg and HBeAg)nin HepG2.2.15 cells. Meanwhile, TA significantly decreased the mRNAnexpression of HBV RNA, which include total HBV RNA, HBV pregenomicnRNA, and HBV precore mRNA. Besides, TA evidently downregulated thenactivity of HBV promoters in HepG2.2.15 cells. Furthermore, we foundnthat TA upregulated the expression of IL-8, TNF-α, IFN-α,nand IFN-α-mediated antiviral effectors in HepG2.2.15 cells.nOn the contrary, TA downregulated the expression of IL-10 and hepaticnnuclear factor 4 (HNF4α). In addition, TA activated the NF-κBnand MAPK pathways that contributed to the inhibition of HBV replication.nFinally, TA treatment led to the occurrence of autophagy, which acceleratednthe elimination of HBV components in HepG2.2.15 cells. Taken together,nour results elucidated the suppressive effect of TA on HBV replicationnand provided inspiration for its clinical application in HBV treatment.","PeriodicalId":522668,"journal":{"name":"Figshare","volume":"1 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2023-07-14","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147901434","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
