Conventional magnetic tumor targeting has used simple staticnmagnetic fields (SMFs), which cause SPIONs to linearly aggregate intona long chain-like shape. Such agglomeration greatly hinders the intracellularntargeting of SPIONs into tumors, thus reducing the therapeutic efficacy.nIn this study, we investigated the enhancement of the intracellularnuptake of SPIONs through the application of rotating magnetic fieldsn(RMFs). Based on the physical principles of SPION chain disassembly,nwe investigated physical parameters to predict the chain length favorablenfor intracellular uptake. Our prediction was validated by clear visualizationnof the intracellular distributions of SPIONs in tumor cells at bothncellular and three-dimensional microtissue levels.nTo identify the potential therapeutic effects of enhanced intracellularnuptake, magnetic hyperthermia as antitumor therapy was investigatednunder varying conditions of magnetic hyperthermia and RMFs. The resultsnshowed that enhanced intracellular uptake reduced magnetic hyperthermiantime and strength as well as particle concentration. The proposednmethod will be useful in the development of techniques to determinenthe optimized physical conditions for the enhanced intracellular uptakenof SPIONs in antitumor therapy.","PeriodicalId":522558,"journal":{"name":"Figshare","volume":"1 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2023-07-21","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147899295","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
