The PEGPD@SDF-1 hydrogel was synthesized from polyethylenenglycol diacrylate (PEG-DA) based scaffolds, dithiothreitol (DTT),nand a novel designed functional peptide module (FPM) via Michael-typenaddition reaction, and the hydrogel was further loaded with stromalncell derived factor-1 (SDF-1). The FPM exhibiting a structure of anchornpeptide–short antimicrobial peptide (SAMP)–anchor peptidencould be cleaved by gingipain specifically, and the SAMP was releasednout of the hydrogel for antibacterial effect in response to gingipain.nThe hydrogel properties were characterized by scanning electron microscopyn(SEM), Fourier transform infrared spectroscopy (FTIR), swelling rationanalysis, degradation evaluation, and release curve description ofnthe SAMP and SDF-1. Results in vitro indicated thenPEGPD@SDF-1 hydrogel exhibited preferable biocompatibility and couldnpromote the proliferation, migration, and osteogenic differentiationnof periodontal ligament stem cells (PDLSCs). Antibacterial testingndemonstrated that the PEGPD@SDF-1 hydrogel released the SAMP stressfullynin response to gingipain stimulation, thereby strongly inhibitingnthe growth of Porphyromonas gingivalis. Furthermore,nthe study in vivo indicated that the PEGPD@SDF-1nhydrogel inhibited P. gingivalis reproduction, createdna low-inflammatory environment, facilitated the recruitment of CD90+/CD34–nstromal cells, and induced osteogenesis. Taken together, these resultsnsuggest that the gingipain-responsive PEGPD@SDF-1 hydrogel could facilitate in situ periodontal tissue regeneration and is a promisingncandidate for the on-demand local drug delivery system for periodontitis.","PeriodicalId":521456,"journal":{"name":"Figshare","volume":"1 1","pages":""},"PeriodicalIF":0.0,"publicationDate":"2021-07-29","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147878797","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
