In the story, several blind men each touch a different part of an elephant—its trunk, leg, ear, or tail—and conclude that the creature resembles a snake, tree, fan, or rope. Each perception is valid within its narrow scope but incomplete when viewed in isolation. Similarly, cancer research has often been fragmented, with investigators focusing on isolated molecular pathways, cellular behaviors, or clinical outcomes without fully connecting these pieces into a unified picture of the disease. This systematic review explores how integrative and multidisciplinary approaches are overcoming that fragmentation to reshape our understanding of cancer. Drawing on advances in systems biology, multi-omics technologies, artificial intelligence, and collaborative research frameworks, we illustrate how convergence across disciplines enables a more comprehensive view of tumor biology and therapeutic response. Major initiatives such as The Cancer Genome Atlas, pan-cancer analyses, and emerging computational platforms exemplify how data integration can reveal patterns invisible to single-dimension studies. By highlighting both the transformative potential and persistent challenges of such integration—ranging from data harmonization to interdisciplinary communication—we propose a roadmap toward a holistic, collaborative, and patient-centered paradigm in oncology. In doing so, we aim to move beyond the limitations of partial understanding toward a collective vision that more accurately reflects the complexity of cancer.","PeriodicalId":515290,"journal":{"name":"Tumor Discovery","volume":"0 0","pages":"025270059-025270059"},"PeriodicalIF":0.0,"publicationDate":"2025-11-21","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147892456","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

Its inhibitory effects on solid tumors have attracted increasing interest and have been actively explored in recent years. This study investigates the therapeutic effects and pharmacological mechanisms of HHT on pancreatic cancer, focusing on mitochondrial energy metabolism and macrophage sensitivity to HHT. HHT significantly inhibited the proliferation and colony formation of pancreatic cancer cell lines PANC-1 and Pan02 in vitro and suppressed tumorigenic potential in vivo. The mechanistic study revealed that HHT induced a significant elevation of reactive oxygen species (ROS) levels in pancreatic cancer cells over time, as evidenced by the enhanced dichlorodihydrofluorescein diacetate fluorescence and an elevated NAD+/NADH ratio. This resulted in mitochondrial respiratory dysfunction, including reductions in basal respiration, maximal respiration, and adenosine triphosphate production. In addition, HHT caused cell cycle arrest and disrupted the cytoskeleton, thereby inhibiting cell division and proliferation. The anti-tumor effects of HHT were further evaluated using a subcutaneous pancreatic tumor-bearing mouse model, showing that HHT inhibited the proliferation of pancreatic tumor cells in vivo, which led to reduced tumor mass. Moreover, HHT significantly reduced the viability of macrophages both in vitro and in vivo, leading to the depletion of tumor-associated macrophages in the tumor microenvironment (TME), thereby alleviating immune suppression. In conclusion, HHT effectively inhibits pancreatic cancer progression through upregulating cellular ROS levels over time, thereby disrupting mitochondrial respiratory capacity in tumor cells and reducing macrophage populations, contributing to TME reprogramming and immune restoration.","PeriodicalId":515290,"journal":{"name":"Tumor Discovery","volume":"4 1","pages":"99-99"},"PeriodicalIF":0.0,"publicationDate":"2025-02-24","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"147921015","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"OA","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

These malformations are strictly confined to the midline, extending from the columella to the glabella. In some cases, they may extend deeply, potentially reaching the leptomeningeal spaces. Imaging studies are crucial for determining the depth of extension and the relationship between the malformation and surrounding structures, such as the cribriform plate and meninges. Detection of frontal and/or nasal bone defects on computed tomography scans or the presence of a transosseous canal with or without a cyst on magnetic resonance imaging should raise suspicion of a dermal sinus cyst prompt surgical excision is indicated to mitigate the risk of the most concerning complication: Neuro-meningeal superinfection. Preferably, surgical excision should be carried out before the age of 1 year to minimize the risk of infection. If communication with meningeal spaces is suspected, neurosurgical intervention becomes necessary. Herein, we present the case of a 4-year-old male child with a history of neonatal distress, psychomotor delays, and epileptic seizures. The patient presented a fistula at the columella level, accompanied by pus discharge, within the context of dysmorphic syndrome. Consequently, the patient was scheduled for fistula excision.","PeriodicalId":515290,"journal":{"name":"Tumor Discovery","volume":" 30","pages":""},"PeriodicalIF":0.0,"publicationDate":"2024-07-04","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"141677921","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
