In oxidative stress (OS) conditions, an imbalance between pro-apoptotic and anti-apoptotic genes can increase the apoptosis rate. In some diseases, such as cancers, patients lose fertility due to chemotherapy and radiotherapy. One way to maintain fertility in these patients is ovarian tissue cryopreservation and transplantation (OTC/T). Studies show that OTC/T also increases apoptosis due to hypoxia and ischemia but fortunately new findings show that applying new approaches could decrease apoptosis in these procedures considerably. This article reviewed follicular atresia, apoptosis during cryopreservation and transplantation, and factors affecting the reduction of apoptosis.","PeriodicalId":495107,"journal":{"name":"Immunoanalysis","volume":"18 10","pages":""},"PeriodicalIF":0.0,"publicationDate":"2024-05-06","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"141010977","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

The mammalian target of rapamycin (mTOR) is one of the regulators for the proliferation and survival of cancer cells. This study focused on the effect of DHA on cellular and exosomal expression of mTOR and related tumor-suppressor microRNAs (miRs) in triple-positive (BT-474) and triple-negative (MDA-MB-231) breast cancer (BC) cell lines. Methods: BT-474 and MDA-MB-231 cells were treated with 100 μM DHA under hypoxic and normoxic conditions for 24 hours. The exosomes were isolated by ultracentrifuge and determined by electron microscopy and CD9, CD63, and CD81 immunoblotting. cDNAs from cellular and exosomal total RNA were used for evaluation of the expression of mTOR and related tumor-suppressor miRs, miR-101 and miR-214, by quantitative real-time PCR. Results: We demonstrated that DHA significantly decreased cellular and exosomal expression of mTOR in both normoxic and hypoxic conditions for both cell lines. Consistently, DHA caused significantly increased expression of miR-214 in all treated groups. However, altered expression of miR-101 showed different patterns in cells and exosomes. Conclusion: According to the beneficial effect of DHA in decreasing the expression of a master regulator for the proliferation of cancer cells, mTOR, in part by increased expression of miR-214, it could be used as a supplementary therapy in BC treatment. Also, miRNA replacement therapy would be useful by suppressing the expression of mTOR in BC treatment.","PeriodicalId":495107,"journal":{"name":"Immunoanalysis","volume":"58 4","pages":""},"PeriodicalIF":0.0,"publicationDate":"2024-01-20","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"140502229","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}

However, its high doses are toxic and may result in a heartbeat, nausea, vomiting, etc. Methods: In the present research, the bimetallic Fe/Mn-MOF modified carbon-ceramic electrode (CCE) was used for studying the electrochemical behavior and determination of nifedipine. The presence of Fe/Mn-MOF on the surface of the CCE as an efficient modifier caused an increase in the current density of nifedipine oxidation. In this way, optimization of some effective parameters consisting of buffer type, pH, modifier concentration, etc. was successfully performed. Moreover, other analytical parameters including reproducibility, repeatability, stability, and selectivity were investigated. Results: The relative standard deviation (RSD) of peak current was 3.2 % for 10 µmol.L-1 nifedipine. Meantime, the linear range of 1-200 µmol.L-1. Additionally, limit of detection (LOD) and limit of quantification were obtained 0.24 μmol.L-1 and 0.82 µmol.L-1 respectively. The results showed that the modified electrode has a good electro-oxidation capability for nifedipine. Finally, for investigating the accuracy of the fabricated sensing assay, the determination of nifedipine in drug and human serum samples was performed. The recoveries and the related RSDs% were obtained 96.4-102% and 2.2-3.6% respectively. Conclusion: This modified electrode exhibited appropriate stability, low cost, simple preparation, fast response time, high repeatability, and good sensitivity. The obtained results clearly demonstrate the applicability of the proposed sensing assay in various matrix effects which does not affect the electrochemical response.","PeriodicalId":495107,"journal":{"name":"Immunoanalysis","volume":"17 1","pages":"0"},"PeriodicalIF":0.0,"publicationDate":"2023-09-17","publicationTypes":"Journal Article","fieldsOfStudy":null,"isOpenAccess":false,"openAccessPdf":"","citationCount":null,"resultStr":null,"platform":"Semanticscholar","paperid":"135303993","PeriodicalName":null,"FirstCategoryId":null,"ListUrlMain":null,"RegionNum":0,"RegionCategory":"","ArticlePicture":[],"TitleCN":null,"AbstractTextCN":null,"PMCID":"","EPubDate":null,"PubModel":null,"JCR":null,"JCRName":null,"Score":null,"Total":0}
