The first protocol adapted for plants was described in the 1990s, however, a universal standardized procedure is not available to date. This study aimed to evaluate the influence of exposure time in the comet assay in plant models Allium cepa L. and Lactuca sativa L. Root tips of both plant models were exposed to methyl methanesulfonate (MMS), a known genotoxicant, for 2 h to 36 h. The alkaline version of the comet assay for plant samples was performed, and the DNA damage was assessed. Arbitrary units (AU) were calculated and compared to the negative control. MMS induced DNA damage (P < 0.05) after 2, 6, 10, 14, 28, 32 and 36 h of exposure in A. cepa, and at 2, 8, 10, 14 and 36 h for L. sativa. The highest AU values for A. cepa were observed after 4 h of exposure to MMS and for L. sativa, after 24 h of exposure. Most of these biocides have short half-lives in water, but little information is available regarding their degradation in sediments. Thus, there is a need to establish a reliable duration of the equilibrium phase for sediment spiking prior to sediment toxicity testing to determine the actual exposure concentrations during ecotoxicological tests. This study aimed to evaluate the degradation of DCOIT, Irgarol, Diuron, and Dichlofluanid during a spiking equilibrium phase of 24 h at three different time intervals (0, 6, and 24 h) and concentrations (10, 100, and 1000 ng g-1), by applying kinetic degradation models. The models presented a better fit for the 1000 ng g-1 treatments, in which the half-lives of DCOIT and Diuron were < 5 h, Dichlofluanid < 2 h, and Irgarol < 6h. Our results also indicate that, except for Dichlofluanid, the degradation rates of the other antifouling biocides were reduced dramatically after 6 h of equilibrium. Therefore, an equilibrium phase of 24 h (or greater than 6 h) was considered viable for sediment spiking. Nostro","doi":"10.5132/eec.2024.01.03","DOIUrl":"https://doi.org/10.5132/eec.2024.01.03","url":null,"abstract":"The agricultural production of the Argentine Pampa is based on intensive cultures, and the most widely used insecticides correspond to cypermethrin (pyrethroid) and chlorpyrifos (organophosphate), among others. Sperm motility and fertility assessments are essential topics for biology and population dynamics studies on fish species. In this study the alterations of these endpoints after exposure to commercial formulation of both insecticides were used to analyze the effects on common carp sperm. After the in vitro exposure at nominal concentrations of 0.25, 0.5, 1, and 2 μg/L of cypermethrin, a significant reduction in motility in 0.5 μg/L concentrations was observed respect control group. The same nominal concentrations were tested for chlorpyrifos, and the same significative effect was detected at 2.0 μg/L. Fertilization rate was significantly reduced at concentrations from 0.5 μg/L cypermethrin exposure, and from 1.0 μg/L in the case of chlorpyrifos. Eggs LC50 / 24 h (95% confidence limit) exposed to cypermethrin was 0.57 μg/L (0.36 - 0.88), and exposed to chlorpyrifos was 2.45 μg/L (1.13-5.29). For the role experiments mini colonies of Zoanthus sp. were exposed to 10 mg/L of PVC microplastics for 7 and 14 days. Exposure time had a direct influence under the PVC microplastics absorption, what consequently contributed for alterations on GST activity, TBARS concentration and DNA damage.
