Abstract
Red pigmentation around the fruit core is a common phenomenon in peaches, resulting from the accumulation of anthocyanins which is always associated with sour taste. However, minimizing this pigmentation is a key breeding objective for developing near-core-uncolored cultivars. To elucidate the mechanism underlying this trait, we compared the near-core-colored cultivar ‘JinXiu’ (JXiu) and the near-core-uncolored cultivar ‘JinXing’ (JXing). The anthocyanin content in JXiu was markedly higher than in JXing, with cyanidin-3-O-glucoside (C3G) identified as the predominant anthocyanin component, showing the greatest difference at the second ripening stage. Integrative analyses of transcriptomic and metabolomic data identified 7,186 differentially expressed genes (DEGs) across ripening stages. Weighted gene co-expression network analysis (WGCNA) revealed that the Magenta module was highly correlated (r > 0.9) with six polyphenols, including C3G. Key anthocyanin biosynthetic genes such as PpPAL2, PpCHS1-3, PpF3H1, PpANS, and Pp4CL6 showed strong co-expression and significantly higher expression in JXiu. Genome-wide variant analysis of JXiu F1 population further identified PpMYB10.1 (Prupe.3G163100) as the most divergent gene between red-cored and non-red-core accessions, with 38 SNPs forming a complete co-segregation across the analyzed individuals. Allele-specific expression analysis revealed predominantly monoallelic transcription of PpMYB10.1 in JXiu, with expression predominantly driven from the reference allele, suggesting the presence of allele-specific regulatory mechanisms. Several intragenic SNPs in PpMYB10.1 (Pp03:18222329, Pp03:18222365, and Pp03:18222368) were linked to allele-specific transcription occurring exclusively at stage r2, coinciding with maximal anthocyanin accumulation. These findings identify PpMYB10.1 as a strong candidate gene associated with anthocyanin accumulation and near-core coloration in peach and provide molecular breeding tool for this horticulturally important trait.
期刊介绍:
Scientia Horticulturae is an international journal publishing research related to horticultural crops. Articles in the journal deal with open or protected production of vegetables, fruits, edible fungi and ornamentals under temperate, subtropical and tropical conditions. Papers in related areas (biochemistry, micropropagation, soil science, plant breeding, plant physiology, phytopathology, etc.) are considered, if they contain information of direct significance to horticulture. Papers on the technical aspects of horticulture (engineering, crop processing, storage, transport etc.) are accepted for publication only if they relate directly to the living product. In the case of plantation crops, those yielding a product that may be used fresh (e.g. tropical vegetables, citrus, bananas, and other fruits) will be considered, while those papers describing the processing of the product (e.g. rubber, tobacco, and quinine) will not. The scope of the journal includes all horticultural crops but does not include speciality crops such as, medicinal crops or forestry crops, such as bamboo. Basic molecular studies without any direct application in horticulture will not be considered for this journal.
