The amount and the quality of the DNA obtained for each one of those methods are variable. The study aimed to evaluate bacterial DNA extraction using conventional boiling method followed by alcohol precipitation . DNA extraction from Gram negative bacilli was extracted and precipitated using boiling method with further precipitation by ethanol. The extraction procedure performed using the boiling method resulted in high DNA yields for both E. coli and K. pneumoniae bacteria in (199.7 and 285.7μg/ml, respectively) which was close to control method (229.3 and 440.3μg/ml). 摘要

脱氧核糖核酸(DNA)的几种提取方法已被应用于细菌DNA的提取。每种方法获得的DNA的数量和质量都是可变的。本研究的目的是评价传统沸煮法后酒精沉淀法提取细菌DNA的效果。采用煮沸法提取革兰氏阴性杆菌DNA,再用乙醇进一步沉淀。沸腾法提取大肠杆菌和肺炎克雷伯菌的DNA提取率分别为199.7和285.7μg/ml,与对照法(229.3和440.3μg/ml)接近。结果表明,酒精沉淀后的沸煮法操作简单,成本低,适用于革兰氏阴性菌DNA的高产质量。
