These dosage forms are often inherently labile due to agglomeration, sedimentation etc., and can be stabilized and manufactured by freeze drying. Lyophilization is the most common method for manufacturing solid pharmaceuticals products and is central to the preservation of materials which must be dried thoroughly in order to ensure stability and require a gentle, easily sterilizable process. Materials and Methods: The current research work involves the formulation of cyclophosphamide injections using lyophilization technique by using different concentrations of cyclophosphamide. The formulated cyclophosphamide injections are further subjected to different evaluation tests such as melting point, differential scanning calorimetry, dissolution rate study, solution stability, pH stability, rubber stopper compatibility and photostability. Results: The moisture content of the formulation is greatly reduced to as low as 0.9% (w/w) thus enhancing the stability of the product. 摘要

导读:对于起效快、生物利用度100%的药物,尤其是口服给药生物利用度差的药物,一般采用肠外给药。新开发的药物往往溶解度差,需要新的剂型,如脂质体、纳米颗粒,以尽量减少溶解度问题和毒性副作用。这些剂型通常由于结块、沉淀等而具有固有的不稳定性,并且可以通过冷冻干燥来稳定和制造。冻干是制造固体药品最常用的方法,是保存必须彻底干燥的材料的核心,以确保稳定性,需要温和,易于灭菌的过程。材料与方法:采用冻干技术制备不同浓度的环磷酰胺注射剂。对配制的环磷酰胺注射剂进行了熔点、差示扫描量热法、溶出率、溶液稳定性、pH稳定性、橡胶塞相容性和光稳定性等评价试验。结果:制剂的水分含量大大降低至0.9% (w/w),提高了产品的稳定性。结论:冻干技术为研制稳定的环磷酰胺注射剂型提供了有利条件。
