摘要

目的探讨激活α7烟碱乙酰胆碱受体(α7n AchR)对氧-葡萄糖剥夺(OGD)所致大脑皮质神经元损伤的影响及其可能机制。方法培养7 d的大脑皮层神经元随机分为3组:对照组、OGD组(氧糖剥夺12 h)和OGD组(PNU-282987预处理24 h,氧糖剥夺12 h),用CCK-8法测定细胞活力,用流式细胞术检测乳酸脱氢酶(LDH)反映细胞损伤、凋亡和活性氧(ROS)的产生,用流式细胞术检测血红素加氧酶-1(HO-1)和缺氧诱导因子-1α(HIF-1α)的表达免疫印迹分析。结果OGD导致细胞死亡,LDH升高,细胞凋亡。与OGD组相比,PNU-282987预处理组细胞存活率显著提高(P 0.05), LDH水平和ROS生成显著降低(P 0.05),细胞凋亡明显抑制(P 0.05)。与OGD组相比,PNU-282987预处理组HO-1蛋白表达显著升高,HIF-1α蛋白表达显著降低(P < 0.05)。结论α7n AchR的激活对ogd诱导的脑皮质神经元损伤具有保护作用,其机制可能与抗氧化应激有关。
