Association of genotypes of SNP c.957A>C with growth using t-test. There were 7 SNPs in exon 2 of the PLAG1 gene, namely SNP c.339A>G, c.489C>T, c.795A>G, c.957A>C, c.1023C>T, c.1056A>G, and c.1353A>G. SNP c.957A>C was validated by PCR-RFLP using TaqI enzyme and obtained three genotypes, namely genotypes AA, AC, and CC with allele frequency A and C, respec-tively 0.10 and 0.90 in Bali cattle, while in PO and Limousine cattle were monomorphic. Genotype association of SNP c.957A>C PLAG1 gene were not associated with birth weight (BW0), weaning weight at 205 days of age (WW205), yearling weight at 365 days of age (YW365), yearling weight at 730 days of age (YW730), and average daily gain (ADG). PLAG1基因是影响牛生长性状的基因之一,位于14号染色体上。本研究旨在获得巴厘牛PLAG1基因外显子1和外显子2的SNP及其与生长性状的关系。样本数量为:巴厘岛牛52个,土生牛10个,豪华牛8个。采用直接测序法鉴定snp的PLAG1基因,采用PCR-RFLP对所选snp进行基因分型。用t检验分析SNP C . 957a >C基因型与生长的关系。PLAG1基因外显子2共有7个SNP,分别为C . 339a >G、C . 489c >T、C . 795a >G、C . 957a >C、C . 1023c >T、C . 1056a >G、C . 1353a >G。利用TaqI酶对SNP C . 957a >C进行PCR-RFLP验证,得到3个基因型,即AA型、AC型和CC型,等位基因频率A和C在巴厘牛中分别为0.10和0.90,而在PO和Limousine牛中为单态。SNP C . 957a >C PLAG1基因的基因型关联与出生体重(BW0)、断奶体重(WW205)、365日龄体重(YW365)、730日龄体重(YW730)和平均日增重(ADG)无相关性。SNP C . 957a >C作为巴厘牛生长性状的候选标记,有待进一步研究。
Identification of single nucleotide polymorphism c.957A>C of PLAG1 gene and its association with growth traits in Bali cattle
