Synthetic Transformations of Higher Terpenoids. 48. Synthesis and Cytotoxic Properties of 18-Nor-Labdatriene-Containing Ureas
A series of 4-ureido-18-nor-15,16-epoxy-8(17),13,14-labdatrienes were synthesized by the reaction of 4-isocyanato-18-nor-lambertianic acid with ammonia, primary and secondary amines, amino acids, and phenylamines. The reactions occurred under mild conditions with high yields of the target products. The cytotoxicity of the synthesized compounds was assessed against cultivated human tumor cells MCF-7 and HEK293T and fibroblasts hTERT-BJ1 (MTT assay). Hierarchical clustering was used for qualitative analysis of the structure–cytotoxicity relationship and revealed several patterns for the effect of the substituent nature on the cytotoxicity and selectivity. The highest selectivity against tumor cells (SI = 2.2–2.3) was found for 3, 5e, and 11b.
