Anthocyanins, as one of the most important active substances in plants, have high nutritional value of human beings. At present, anthocyanin biosynthesis has been widely studied, however, the regulatory mechanism of anthocyanins in <em>Dendrobium officinale</em> (<em>D. officinale</em>) remain unknow. Here, the anthocyanin components of two different colored cultivars from <em>D. officinale</em> were analyzed by UPLC-MS/MS. We have found that delphinidin and its derivatives were the main anthocyanin components in <em>D. officinale</em>, among which Malvidin-3-O-glucoside and Malvidin-3-O-galactoside were the main active substances. Subsequently, the key structural genes <em>DoCHS1/2</em>, <em>DoF3H</em>, <em>DoF3’H</em>, <em>DoDFR1/2</em>, <em>DoANS</em>, and transcription factors <em>DoMYB5–1</em>, <em>DoMYB7</em>, <em>DobHLH5–1</em> were identified based on transcriptomic analysis, which were mainly involved in the anthocyanin biosynthesis. Among that, both <em>DoMYB5–1</em> and <em>DoMYB7</em> could interact with <em>DobHLH5–1</em> to form complexes (<em>DoMYB5–1-DobHLH5–1</em>, <em>DoMYB7-DobHLH5–1</em>), and then regulated the expression of <em>DoDFR1</em> and <em>DoF3’H</em> to promote anthocyanin synthesis through the protein-protein interaction, transient overexpression experiments and Y1H assay. Furthermore, we also found that the interaction of <em>DoMYB5–1</em> and <em>DobHLH5–1</em> could cascade-rergulate the complex <em>DoMYB7-DobHLH5–1</em>, thereby regulating the expression of <em>DoDFR1</em> and then participating in the regulation of anthocyanin metabolism in <em>D. officinale</em>. 摘要
MYB和bHLH转录因子可能通过相互作用或上下游调控参与花青素的合成。花青素作为植物中最重要的活性物质之一,具有很高的人体营养价值。目前,花青素的生物合成已被广泛研究,但铁皮石斛中花青素的调控机制尚不清楚。采用超高效液相色谱-质谱联用技术对两个不同花色品种的花色苷成分进行了分析。我们发现,海鸽苷及其衍生物是officinale中花青素的主要成分,其中Malvidin-3-O-glucoside和malvidin -3- o -半乳糖苷是主要活性物质。随后,通过转录组学分析鉴定出主要参与花青素生物合成的关键结构基因DoCHS1/2、DoF3H、DoF3'H、DoDFR1/2、DoANS和转录因子DoMYB5-1、DoMYB7、DobHLH5-1。其中,DoMYB5-1和DoMYB7均可与DobHLH5-1相互作用形成复合物(DoMYB5-1 - DobHLH5-1、DoMYB7- DobHLH5-1),通过蛋白相互作用、瞬时过表达实验和Y1H测定,调控DoDFR1和DoF3'H的表达,促进花青素合成。此外,我们还发现DoMYB5-1和DobHLH5-1的相互作用可以级联调节DoMYB7-DobHLH5-1复合物,从而调节DoDFR1的表达,进而参与调节officinale花青素代谢。综上所述,本研究明确了铁皮石斛花青素积累的调控机制,有助于揭示植物次生代谢的生物学机制,为铁皮石斛品质改良提供理论参考。
