PM2.5对GDM的总影响由g-Phascolarctobacterium(1.83 %)、s-Phascolarctobacterium_unclassified(1.83 %)和s-Bacteroides_uniformis(0.65 %)介导。

The mediating role of gut microbiota in the influence of PM2.5 exposure on gestational diabetes mellitus

Objective

Our study aimed to explore the mediating effects of the gut microbiota on the associations of PM2.5 exposure with gestational diabetes mellitus (GDM).

Methods

Participants (n = 1268) were recruited in Guangzhou, China. The participants’ information and fecal samples were collected, the air pollutants exposures during pregnancy was assessed using measurements from the nearest monitoring station, and the relative abundance of fecal microbiota was characterized by 16S rRNA sequencing. Linear discriminant, mediation, and regression analyses were conducted to estimate the associations among PM2.5 exposure, gut microbiota and GDM.

Results

Elevated PM2.5 exposure was significantly correlated with the alterations in the gut microbiota, and with the increased risks of GDM and impaired glucose homeostasis. The mediation analyses indicated that alterations in the gut microbiota partially mediated the effects of PM2.5 exposure on GDM and impaired glucose homeostasis. The total effects of PM2.5 on GDM were mediated by g-Phascolarctobacterium (1.83 %), s-Phascolarctobacterium_unclassified (1.83 %), and s-Bacteroides_uniformis (0.65 %). The corresponding mediating effects for increased fasting glucose level were 3.48 %, 8.55 %, 1.76 %, 5.14 % and 0.02 % by g-Bacteroides, g-Lachnospiraceae_Ruminococcus, g-Streptococcus, g-Dorea and s-Bacteroides_uniformis, respectively.

Conclusions

Elevated PM2.5 exposure from 3 months before pregnancy to the 12th week of gestation may induce GDM via alteration of the gut microbiota.
