Pseudidiomarina salilacus sp. nov., a novel bacterium of the genus Pseudidiomarina isolated from sediment in Yuncheng Salt Lake.
Three Gram-stain-negative, aerobic, rod-shaped, yellow-pigmented bacteria, designated YC-516-91T, YC-516-48 and YC-516-93, were isolated from a sediment sample of Yuncheng Salt Lake in China. Growth occurred at 15-42 ℃ (optimum 32-37 ℃), pH 6.0-10.0 (optimum pH 7.0-8.0) and with 1-11.0% NaCl (w/v, optimum 3%). Strain YC-516-91T was positive for hydrolysis of Tween 80 and starch, but negative for nitrate reduction, urease, H2S production, hydrolysis of Tween 40 or gelatin. Phylogenetic analysis based on 16S rRNA gene and genome sequences showed that YC-516-91T, YC-516-48 and YC-516-93 belonged to the genus Pseudidiomarina. 16S rRNA gene sequence similarity values of YC-516-91T to the closest related type strains Pseudidiomarina atlantica MCCC T A10513T, Pseudidiomarina terrestris 1APP75-27aT and Pseudidiomarina donghaiensis 908033T were below 97.99%. Strains YC-516-91T, YC-516-48 and YC-516-93 have average nucleotide identity (ANI) values of 99-100%, digital DNA-DNA hybridization values of 92.1-97.4% and average amino acid (AAI) values of 99-100%, suggesting that they belong to the same species. These strains are closely related to P. atlantica MCCC 1A10513T, with ANI and AAI of around 92-93%, below the species delineation cut-off, thus indicating that they represent a novel species. The genome of strain YC-516-91T was 2,723,173 bp, and the genomic DNA G+C content was 50.24 mol%. The only respiratory quinone was Q-8, and the major fatty acids (>10%) were iso-C17 : 0 and iso-C15 : 0. The major polar lipids consisted of diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylglycerol, phospholipid, glycolipid, unidentified aminolipids and two unidentified polar lipids. Thus, strains YC-516-91T, YC-516-48 and YC-516-93 represent a novel species based on their chemotaxonomic, phenotypic and phylogenetic characteristics. Strain YC-516-91T is proposed to represent a novel species named Pseudidiomarina salilacus sp. nov., within the genus Pseudidiomarina. The type strain is YC-516-91T (=MCCC 1K09529T=KCTC 8778T).
