Recent advances in structural biology, including cryo-electron microscopy and computational tools like AlphaFold, have unveiled the structural intricacies and diversity of these proteinaceous layers. This review highlights the biogenesis, structural assembly, evolutionary adaptations, and functional roles of archaeal S-layers, focusing on model organisms such as <em>Sulfolobus acidocaldarius</em> and <em>Haloferax volcanii</em>.
