In this work, we identified that the BFSs are primarily caused by missing quenchers in some TP constructs and surface-adsorbed dye-labeled DNA strands. To address these issues, we developed a double-quencher TP (dqTP) and incorporated Tween-20 treatment during surface preparation. These two simple strategies reduced the BFS level by tenfold, significantly improving the signal-to-background ratio for single molecular force imaging. We demonstrated the performance of dqTP by monitoring the temporal dynamics of integrin tensions in platelets and HeLa cells, showing that single integrin tensions remain stable for at least 100 seconds in wild-type HeLa cells. In contrast, with vinculin knocked out, a subpopulation of integrin tensions, especially at cell peripheral region, exhibited molecular force fluctuations with an average force duration shorter than 10 seconds. BJ welcomes experimental studies that employ quantitative physical approaches for the study of biological systems, including or spanning scales from molecule to whole organism. Experimental studies of a purely descriptive or phenomenological nature, with no theoretical or mechanistic underpinning, are not appropriate for publication in BJ. Theoretical studies should offer new insights into the understanding ofexperimental results or suggest new experimentally testable hypotheses. Articles reporting significant methodological or technological advances, which have potential to open new areas of biophysical investigation, are also suitable for publication in BJ. Papers describing improvements in accuracy or speed of existing methods or extra detail within methods described previously are not suitable for BJ.
