One hundred twenty male Fischer 344 rats were divided into 6 groups. Sixty rats (groups 1, 2 and 3) were given 0.05% BBN in drinking water for the initial 8 weeks of the experiment and another 60 rats (groups 4, 5 and 6) served as controls. HCFU mixed in the powder diet was administered at daily doses of 100 mg per kg of body weight. Group 2 received HCFU throughout and group 3 after BBN administration for 8 weeks. All animals were sacrificed at 20 weeks from the beginning of the experiment and examined histopathologically. In groups 1, 2 and 3, urinary bladder tumors developed in 13 of 18, 0 of 20, and 4 of 19, respectively. Incidences of tumors in the 2 groups treated with HCFU were significantly lower than that of group 1 treated with BBN alone. 本研究探讨了1-己基氨基甲酰-5-氟尿嘧啶(HCFU)对n -丁基- n -(4-羟基丁基)亚硝胺(BBN)致大鼠膀胱肿瘤发展的影响。120只雄性Fischer 344大鼠分为6组。实验前8周,60只大鼠(1、2、3组)灌胃0.05% BBN,另60只大鼠(4、5、6组)作为对照组。将HCFU混合在粉状日粮中,每日剂量为每公斤体重100毫克。第2组在治疗期间给予HCFU治疗,第3组在给药8周后给予BBN治疗。实验开始后20周处死大鼠,进行组织病理学检查。在1、2和3组中,18人中有13人发生膀胱肿瘤,20人中有0人发生膀胱肿瘤,19人中有4人发生膀胱肿瘤。两组患者的肿瘤发生率均明显低于单用BBN治疗的1组。提示HCFU具有抑制膀胱肿瘤发生的作用。
