In recent years, there has been a growing emphasis on genetic engineering of Aspergillus to increase its yield of organic acids. This review provides a comprehensive overview of the current advancement and future directions in the application of genetic engineering techniques to enhance organic production in Aspergillus, specifically highlighting achievement in reconstructing metabolic pathways for desired products, eliminating by-products, modifying regulatory pathways, and engineering mycelial morphology. Furthermore, this review also focuses on the strategies and genetic tools applied in Aspergillus, with particular emphasis on the potential applications and challenges of CRISPR-based biosensors in organic acid fermentation.
