The rats of both sexes (n=17), average weight of 184g were randomly assigned into treatment (n=10) and control (n=7) groups. The rats in the treatment group received 2mg/kg body weight of chloroquine phosphate dissolved in distilled water daily for fourteen days. The control rats received equal volume of distilled water daily. The rats were fed with rat pellets and given water liberally and were then sacrificed on day fifteen of the experiment. The inferior colliculus was carefully dissected out and quickly fixed in 10% formal saline for histological study.\nHistological findings of the nissl substance indicate that the treatment groups showed sparsely distributed neurons, which stain less intensely when compared with the control. The nissl substance in some of the neurons appeared degenerative while some hypertrophied. These findings indicate that chronic administration of chloroquine has a deleterious effect on the nissl substance of the inferior colliculus. Chloroquine may have adverse effects on auditory sensibilities by its deleterious effects on the nissl substance of the inferior colliculus of the adult wistar rats. 摘要
研究了亚急性给药氯喹对成年wistar大鼠下丘(IC)的影响。氯喹常用于预防或治疗疟疾、类风湿关节炎和红斑狼疮。选取平均体重184g的雄性和雌性大鼠17只,随机分为治疗组(n=10)和对照组(n=7)。治疗组大鼠每日给药2mg/kg体重的磷酸氯喹蒸馏水,连续14 d。对照组每天给予等量蒸馏水。实验第15天,用大鼠颗粒喂养大鼠,并大量饮水。仔细解剖下丘,并迅速固定在10%正规生理盐水中进行组织学研究。nissl物质的组织学结果表明,处理组神经元分布稀疏,染色强度低于对照组。部分神经元内的尼索物质退行性变,部分神经元内的尼索物质肥大。这些结果表明,长期服用氯喹对下丘的尼索物质有有害影响。氯喹可能通过对成年wistar大鼠下丘尼索物质的破坏作用而对听觉产生不良影响。建议进行旨在证实这些观察结果的进一步研究。关键词:氯喹,尼氏物质,下丘,wistar大鼠。生物医学科学年鉴Vol.3 (1&2) 2004: pp. 33-38
