摘要

目的:建立聚合酶链反应-对抗性双对引物(PCR-CTPP)对越南人MTHFR rs1801133基因多态性进行分型的方法。方法:采用15名越南人的DNA样本对该方法进行评价。采用Sanger测序法鉴定rs1801133多态性的标准基因型。利用生物信息学软件设计了4个多重PCR-CTPP。进行了实验程序来验证PCR方法的引物和成分。结果:采用最佳内/外引物比对、引物熔融温度、组份及热循环方法,成功建立了MTHFR rs1801133多态性鉴定的PCR-CTPP方案。结论:应用pcr - cttp方法对越南人群MTHFR rs1801133多态性进行大队列基因分型,探讨多态性与代谢综合征的关系。
