摘要

背景甲状腺相关性眼病(TAO)以眼眶脂肪组织增多为特征。瘦素是一种特异性脂肪细胞因子,可能在TAO发病过程中起重要作用。目的探讨瘦素对甲状腺相关性眼病(TAO)患者眼眶前脂肪细胞分化的影响。方法对7例TAO患者行眶减压术,取眶脂肪组织。采用外植体法培养眼眶前脂肪细胞,油红染色鉴定分化细胞。将传代培养的前脂肪细胞与不同浓度的瘦素孵育,对照组不添加瘦素。采用油红染色法测定分化脂肪细胞细胞质/核仁的平均光密度,探讨不同浓度的瘦素对眼眶脂肪细胞脂质形成的影响。结果体外培养的前脂肪细胞经免疫组化染色鉴定为间充质来源。对照组、10μg/L瘦素组、50μg/L瘦素组、100μg/L瘦素组和500μg/L瘦素组的细胞质/核仁平均光密度分别为1.07±0.22、0.80±0.17、0.56±0.11、0.25±0.10和0.17±0.08,组间差异有统计学意义(F=20.64,P=0.00)。与对照组相比,不同浓度rh-瘦素组眼眶脂肪细胞脂质形成逐渐下降(P<0.05,P<0.01)。结论rh -瘦素可抑制体外TAO眶前脂肪细胞的分化和脂质形成,且呈剂量依赖性。瘦素可能是TAO发病机制中的反馈调节剂。关键词:甲状腺相关性眼病;瘦素;preadipocyte;细胞分化
