TNF α and myeloperoxidase (MPO) in fetal kidney tissue were detected by ELISA and biochemically respectively, and the renal histopathological changes were also observed.Results The activity of TNT α increased gradually with the extension of ischemia, especially after 35~45 minutes (P 0.05) .It rose progressively during reperfusion and was at its highest after 8 hours (P0.01) .It reached the level of the sham operation group after 30 hours reperfusion .The activity of MPO changed as that of TNF α . The activity of MPO reached the peak in 15 hours of reperfusion (P0.01) . 肿瘤坏死因子- α对胎鼠肾急性缺血后炎症反应的影响
目的探讨肿瘤坏死因子α (TNF α)对大鼠胎肾急性缺血后炎症反应的影响。方法采用夹持Wistar大鼠子宫供血侧血管的方法,建立不同程度的子宫急性缺血模型和胎鼠再灌注模型。采用ELISA法和生化法分别检测胎鼠肾组织中TNF α和髓过氧化物酶(MPO)的含量,并观察肾组织病理变化。结果大鼠心肌组织中TNT α活性随缺血时间延长而逐渐升高,尤其是在缺血35~45 min后(p0.05),再灌注时逐渐升高,8 h后达到最高值(P0.01),再灌注30 h后达到假手术组水平,MPO活性随TNF α变化而变化。MPO活性在再灌注15h时达到峰值(P0.01)。再灌注15h后组织学改变最为显著,包括血管充血、小管空泡变性或坏死、中性粒细胞积聚、基膜破裂,尤以小管近端为明显。结论子宫缺血再灌注肾损伤存在炎症反应,TNF α可能是其中重要的介质之一,MPO能反映炎症反应的程度。
