Islets were then isolated by collagenase digestion, purified by continuous gradient Ficoll centrifugation, and islet equivalent (IE) yield determined by dithizone staining. Viability of isolated islets was assayed by using the fluorescent SYTO/ethidium bromide (SYTO/EB) method.
