The emergence of organoid culture system and its application in cancer research provides a unique opportunity to develop novel <em>in vitro</em> cancer pre-clinical models. Here we review the recent advances in utilizing organoids culture system and other related three-dimensional culture systems in studying cancer biology, performing drug screening, and developing cancer therapies.
