Abstract
Background: Psychological stress and health behaviors are associated with oxidative status and telomere biology, but evidence from multidomain interventions in breast cancer survivors remains limited.
Objective: To evaluate the effects of the Pythagorean Self-Awareness Intervention (PSAI) on biological, behavioral, and psychosocial outcomes following breast cancer treatment.
Methods: In this randomized controlled trial, 120 women were randomized to an eight-week PSAI program or education-only control condition (n = 60 each); 118 were included in the modified intention-to-treat analysis. Primary biological outcomes were serum total oxidant status and PBMC relative telomere T/S ratio. Linear mixed-effects models estimated group × time effects, between-group differences in change, 95% confidence intervals (CIs), and Hedges' g.
Results: Total oxidant status decreased more in the intervention group (difference in change, -3.83 μmol/L; 95% CI -7.03 to -0.63; p = 0.020; g = -0.13). Telomere-length change did not differ significantly between groups (-0.170log₂[T/S] units; 95% CI -0.554 to 0.214; p = 0.382; g = -0.23). PSAI also improved physical activity, body weight, sleep, perceived stress, anxiety, and depression.
Conclusion: PSAI improved total oxidant status and several patient-relevant outcomes but did not significantly affect telomere-length change. Longer, adequately powered trials with repeated biomarker assessments are require.
Mechanisms of Ageing and Development is a multidisciplinary journal aimed at revealing the molecular, biochemical and biological mechanisms that underlie the processes of aging and development in various species as well as of age-associated diseases. Emphasis is placed on investigations that delineate the contribution of macromolecular damage and cytotoxicity, genetic programs, epigenetics and genetic instability, mitochondrial function, alterations of metabolism and innovative anti-aging approaches. For all of the mentioned studies it is necessary to address the underlying mechanisms.
Mechanisms of Ageing and Development publishes original research, review and mini-review articles. The journal also publishes Special Issues that focus on emerging research areas. Special issues may include all types of articles following peered review. Proposals should be sent directly to the Editor-in-Chief.
