Abstract
Objective: To investigate in vitro bioactivity of the bioceramic root canal sealer Bioseal.
Methods: An in vitro mineralization model consisting of root canal sealer specimens and simulated body fluid (SBF) was constructed. Bioceramic-based root canal sealers Bioseal, iRoot SP, and silicone root canal sealer GuttaFlow2 were each prepared into cylindrical specimens with a diameter of 10 mm and a height of 2 mm. All the root canal sealer specimens were then immersed in SBF and maintained at 37 ℃ in a water bath for 4 weeks. Scanning electron microscopy (SEM) and X-ray energy dispersive spectroscopy (EDS) were used to observe the morphology of the root canal sealer specimens after immersion. The surface composition was analyzed by X-ray diffraction (XRD). Calcium ion concentration in the extraction solution was measured using inductively coupled plasma optical emission spectrometry (ICP-OES) and pH value was monitored for 1, 2, 7, 14, 21, and 28 days. Statistical analysis was performed using one-way ANOVA test. The level of significance was set at P < 0.05.
Results: After 4 weeks of immersion in SBF, spherical particles were observed on the surfaces of both Bioseal and iRoot SP under SEM. XRD analysis confirmed that the calcium-to-phosphorus ratios were 1.64 and 1.56, respectively, indicating the formation of hydroxyapatite. The calcium ion concentration in bioceramic-based root canal sealers Bioseal and iRoot SP extraction solutions peaked at (110.4±2.8) mg/L and (125.4±3.8) mg/L after 7 days, respectively (P < 0.05), then stabilized (P>0.05). After 2 days, the pH value peaked at 7.99±0.11 and 8.23±0.10, respectively (P < 0.05). The calcium ion concentration and pH value of Bioseal extraction solution were lower than those of iRoot SP at all time points (P < 0.05). No hydroxyapatite formation was observed on the sample surface of silicone root canal sealer GuttaFlow2, and during the evaluation period of 28 days, there were no significant changes in calcium ion concentration or pH value (P>0.05).
Conclusion: The bioceramic root canal sealer Bioseal and iRoot SP demonstrated bioactivity by promoting calcium ion deposition and forming hydroxyapatite in physiological solutions.
