Abstract

To overcome the thermodynamic barrier of “NADH-dependent CO2 reduction”, we developed a bioelectrosynthesis platform via the co-immobilization of CO2 reductase (CR) and diaphorase (DP) on a graphite electrode. A strategic protein engineering approach, utilizing carbon-specific solid-binding peptides (SBPs), enabled oriented enzyme assembly to facilitate efficient interfacial electron transfer. Among the variants, the dual-functionalized system—comprising C-terminal SBP-fused CR and N-terminal ferredoxin (Fd)/SBP-fused DP3—exhibited superior bioelectrocatalytic synergy, achieving a current density of –193.35 µA cm–2. At –0.5 V (vs Ag/AgCl), this integrated interface effectively drove the endergonic reduction process, maintaining continuous cofactor regeneration with an electron transfer efficiency of 39.2%.

ACS Applied Materials & Interfaces is a leading interdisciplinary journal that brings together chemists, engineers, physicists, and biologists to explore the development and utilization of newly-discovered materials and interfacial processes for specific applications. Our journal has experienced remarkable growth since its establishment in 2009, both in terms of the number of articles published and the impact of the research showcased. We are proud to foster a truly global community, with the majority of published articles originating from outside the United States, reflecting the rapid growth of applied research worldwide.
