Abstract
Purpose: Process Analytical Technology (PAT) tools are increasingly applied to improve monitoring of biopharmaceutical processes; however, few techniques have explored their use in liquid bioprocesses based on insect cell cultures. This work evaluated whether rheological parameters could be explored as monitoring indicators for biochemical variables during virus-like particle (VLP) production using the insect cell-baculovirus expression system.
Methods: Batch bioreactor cultivations of Spodoptera frugiperda (Sf9) cells were conducted under non-infected conditions and during rabies and SARS-CoV-2 VLP production. Rheological properties were determined by steady-shear cone-plate rheometry and correlated with viable and total cell densities, extracellular protein concentration, and viral titer. Empirical models were generated and ranked using Akaike's Information Criterion, and predictive performance was assessed by root mean square error. Statistical significance was evaluated using analysis of variance.
Results: All cultures exhibited non-Newtonian, pseudoplastic behavior, with decreases in the flow behavior index associated with increasing cell density and protein concentration. Exponential functions were most frequently selected among the best-ranked empirical models, followed by rational, hyperbolic, logarithmic, and power equations. No statistically significant effect of rheological parameter or infection condition on prediction accuracy was observed (p > 0.05).
Conclusion: Rheological properties reflected compositional changes occurring during insect cell cultivations; however, under the conditions studied, rheology did not significantly improve quantitative monitoring of key biochemical parameters using other approaches. These findings indicate that while rheology may provide complementary process information, further investigation is required before its application as a robust PAT tool for VLP production.
期刊介绍:
Biotechnology Letters is the world’s leading rapid-publication primary journal dedicated to biotechnology as a whole – that is to topics relating to actual or potential applications of biological reactions affected by microbial, plant or animal cells and biocatalysts derived from them.
All relevant aspects of molecular biology, genetics and cell biochemistry, of process and reactor design, of pre- and post-treatment steps, and of manufacturing or service operations are therefore included.
Contributions from industrial and academic laboratories are equally welcome. We also welcome contributions covering biotechnological aspects of regenerative medicine and biomaterials and also cancer biotechnology. Criteria for the acceptance of papers relate to our aim of publishing useful and informative results that will be of value to other workers in related fields.
The emphasis is very much on novelty and immediacy in order to justify rapid publication of authors’ results. It should be noted, however, that we do not normally publish papers (but this is not absolute) that deal with unidentified consortia of microorganisms (e.g. as in activated sludge) as these results may not be easily reproducible in other laboratories.
Papers describing the isolation and identification of microorganisms are not regarded as appropriate but such information can be appended as supporting information to a paper. Papers dealing with simple process development are usually considered to lack sufficient novelty or interest to warrant publication.
