Abstract
Light transmission aggregometry is a method that measures the increase in light transmission of a platelet-rich plasma (PRP) stirred at 37°C after the addition of an agonist. It is used to perform various biological tests, including the in vitro assessment of platelet function for the investigation of bleeding disorders. This method has specific characteristics related to the nature of the matrix used (platelet-rich plasma, PRP) that must be considered for accreditation according to the NF EN ISO 15189 standard. Following a survey of practices conducted among members of the Societe Francaise de Thrombose et d’Hemostase (SFTH), the society’s quality group proposed a synthesis of the various current recommendations and formulated 28 proposals covering pre-analytical, analytical, and post-analytical aspects for the accreditation of tests using optical aggregometry.
These proposals were approved by the Reference Center for Inherited Platelet Disorders and by the members of the SFTH following a vote. They are intended to provide guidance on the production of method validation files for the investigation of platelet functions in PRP in the context of platelet function disorder screening and for the RIPA (Ristocetin-induced platelet aggregation) test. Most of them can nevertheless be used for all other tests that employ this method (e.g., functional tests for the diagnosis of heparin-induced thrombocytopenia). This text is a summary of these proposals.
