Abstract
This study developed three DNA-based PCR assays targeting the cytochrome c oxidase subunit I (COI) mitochondrial gene to identify Ibacus novemdentatus, a smooth fan lobster. Conventional PCR and real-time PCR (RT-PCR) assay exhibited specificity for the smooth fan lobster, with no cross-reactivity with eight non-target species. The assays were further refined to develop an ultrafast RT-PCR assay capable of detecting smooth fan lobster within 30-min PCR reaction time. The specificity, sensitivity, and false-positive and false-negative rates of the three assays complied with the CODEX and US FDA guidelines. We evaluated the applicability of these assays using 17 commercial products. Furthermore, direct lysis buffer (DLB)-based ultrafast RT-PCR has been confirmed as a useful assay for field applications. The three proposed assays are useful tools for accurate detection of smooth fan lobsters in food and have potential applications in the food industry.

Food Analytical Methods publishes original articles, review articles, and notes on novel and/or state-of-the-art analytical methods or issues to be solved, as well as significant improvements or interesting applications to existing methods. These include analytical technology and methodology for food microbial contaminants, food chemistry and toxicology, food quality, food authenticity and food traceability. The journal covers fundamental and specific aspects of the development, optimization, and practical implementation in routine laboratories, and validation of food analytical methods for the monitoring of food safety and quality.
