Abstract
Trans-arterial chemoembolization (TACE), a first-line treatment for intermediate-/advanced-stage hepatocellular carcinoma (HCC), is heavily limited by tumor vascular heterogeneity and poor drug penetration. In this study, a temperature/radiofrequency (RF)-responsive poly(ionic liquid) nanogel (poly(N-isopropylacrylamide-b-1-butyl-3-vinylimidazolium combretastatin A4 phosphate), PIV·CA) was designed to achieve thermo-boosting trans-arterial vascular disruption-embolization (TAVDE) via RF-triggered release/penetration promotion of combretastatin A4 phosphate (CA4P) and temperature-sensitive sol-gel phase transition. The RF field shockingly increased the cytotoxicity of PIV·CA by approximately 22-fold, increasing the interaction of CA4P with tubulin and resulting in cell cycle arrest in the G2/M phase via increased suppression of tubulin polymerization. PIV·CA precisely synergizes vascular disruption, arterial embolization and RF ablation, suggesting a potential strategy of RF-energized TAVDE in transarterial therapy.
